Protective effect of medroxyprogesterone acetate plus testosterone against radiation-induced damage to the reproductive function of male rats and their offspring.

Abstract: This study attempted to protect spermatogenesis and the reproductive performance of rats against the effects of acute scrotal exposure to x-rays. Daily subcutaneous hnjections of medroxyprogesterone acetate (8 mg/kg) plus testosterone (1 mg/kg) (MT group) were administered for 55 days (experiment A) or 15 days (experiment B). The rats were irradiated (3 grays) on the last day of MT pretreatment (MTX group). In both experiments, on days 1 and 130 posttreatment, rats from each of the four groups (control, x-irra… Show more

“…In one study [17], the fertility in rats which received pro carbazine alone was impaired, effects upon both the sperm cell spermatogonia and the developing germs cells being implicated. However, 100 days after the end of cytotoxic/ steroid treatment the testicular size (an indicator of sperm content) was partially preserved, although this was not sig nificant.…”

“…These studies have drawn from the enormous effort being made to develop a male contra ceptive which presently favours the use of steroid to inhibit spermatogenesis indirectly by the withdrawal of gonadotro phin support. In studies using both procarbazine [17] and radiation [18] as the toxic insult, testicular recrudescence was produced by daily injections of medroxyprogesterone acetate and testosterone (MPA-T) which were adminis tered for several weeks before cytotoxic treatment. The suggested high efficiency of this approach contrasts with the very limited or no success with androgens or oestrogen reported previously [4,5,19], The effects of the steroidal contraceptive regimen at the time of cytotoxic treatment were clear: the rats were infertile, epididymal sperm con tents were reduced and the testis was atrophied.…”
