Protective effect of hypothermia and vitamin E on spermatogenic function after reduction of testicular torsion in rats

Abstract: This study was designed to investigate the protective effect of hypothermia and vitamin E on spermatogenic function after reduction of testicular torsion in rats. Ninety-six pure inbred male SD rats were divided into group A, B, C and D according to the principle of body weight and birth similarity, with 24 rats in each group. Four groups of rats were respectively twisted on the left testis to establish unilateral testicular torsion rats. Rats in groups A, B, C, D were respectively given normal saline, hypothe… Show more

“…In the current study; the injection with V and C induced a decline in serum Ts and DHEA-SO 4 levels in the rats, where the major part of male reproductive system is dependent on Ts; the process of spermatogenesis will eventually cease where vitamin E support in the absence of this hormone (Anvari et al, 2020;Sadeghi et al, 2020;Bo et al, 2020). The decrease of Ts may be attributed to: V and C inhibit human chronic gonadotropin-stimulated Ts biosynthesis by both cultured rat precursor and immature leydig cells (Pasqualotto et al, 2003); or the effect of V and C, which localized between enzymatic steps of Ts and DHEA-SO 4 biosynthesis, changing cholesterol to androstenedione through inhibiting 17a-hydroxylase, cholesterol side chain cleavage and 3b HSD-isomerase steps (which change cholesterol to pregnenolone and pregnenolone to progesterone, respectively (Saradha and Mathur, 2006).…”

“…The effect of vitamin E might be through the scavenging of reactive oxygen species induced by V and C (Diemer et al, 2003). Vitamin E reverses testicular injury in rats and declines the apoptosis rate of spermatogenic cells (Bo et al, 2020). Also, vitamin E prevents coal burning fluorosis-induced spermatogenic cell apoptosis through the suppression of oxidative stress (Tian et al, 2018).…”

“…Induction of testicular torsion was carried out according to the model of testicular torsion rats developed by Ranade et al (2011) and Bo et al (2020). After the grouping, rats in the NC group underwent sham/placebo surgery, while those in the PC, T1, and T2 groups were anesthetized with a ketamine injection (Ilium, NSW, 100 mg ketamine/mL) at a dose of 10 mg/kg BW and xylazine (Ilium, NSW, 20 mg xylazine/mL) dose of 2 mg/kg BW.…”
