Protective effect of ghrelin on testicular damages caused by chronic hypoxia in rats: A histopathological study

Abstract: Hypoxia can lead to changes in the blood flow, nutrition and oxygenation of male germ cells and results in fertility reduction through the increase in oxidative stress. This study aims to evaluate the effect of ghrelin on testicular damage induced by hypoxia in rats. In this experimental study, 24 male rats were randomly divided into four groups: control, hypoxia, hypoxia + ghrelin and ghrelin. Animals in the control and ghrelin groups were kept in room air with 21% oxygen. The animals in the groups of hypoxia… Show more

“…It has been established that obestatin plays a role in increasing cell survival and proliferation and counteracting apoptotic process and inflammation [41]. Regarding the reproductive system, a protective activity of ghrelin was observed in different researches, performed in animal models, demonstrating that this peptide can decrease the testicular damages [17, 4244]. Recent studies deal with the defensive role of ghrelin that can exert beneficial activity on redox imbalance for its anti-inflammatory and antioxidant properties also in the male reproductive system [18, 45–47].…”

“…PAS staining was used to evaluate glycogen content in (glycogen storage) in hepatocytes. Finally, to evaluate the tissue changes in each sample, 12 images were randomly selected then the histological index was calculated using semiquantitative method described by Frei et al The level of damage was recorded based on (0-3) grades in which grade 0 = no damage, 1 = mild damage, 2 = moderatedamage, and 3 = severe damage (20)(21)(22)(23).Slides were viewed and photographed using a camera microscope at 400x magnification in at least three random microscopic fields from each animal by two expert pathologists without knowledge of the treatment groups [22][23][24][25].…”

“…The morphometric analysis was performed to calculate seminiferous tubules' diameter and the height of the germinal epithelium (HE) in seminiferous tubules. Besides, the thickness of the basement membrane of seminiferous tubules was measured using PAS staining (periodic acid-Schiff) (Hazrati, Salimnejad, Alipour, Mirzaei Bavil, & Alihemmati, 2018). The histopathological alterations were assessed semi-quantitatively with the use of a four-point scale:…”
