Protective effect of decorin on acute ischaemia-reperfusion injury in the rat kidney

Abstract: IntroductionTransforming growth factor-β1 (TGF-β1) has a crucial role in collagen synthesis and fibrosis. TGF-β1 can be antagonized and/or reduced by the action of certain agents. We propose to identify the role of decorin in treatment of tubular and interstitial fibrosis and in the inhibition of TGF-β1 in an acute ischaemic kidney.Material and methodsWe grouped 34 female Sprague Dawley type rats into 3 groups as 9 sham, 9 ischaemia-reperfusion (I/R) and 16 I/R + decorin respectively. The rats in the I/R + dec… Show more

“…Based on the abovementioned knowledge, therapeutic approaches that involve blocking the pathogenic action of TGF-β on multiple levels have been developed to control the progression of renal fibrosis. Preclinical research on some of these promising interventions has been undertaken383940; however, few studies have progressed to clinical trials. Unacceptable adverse effects induced by complete TGF-β inhibition were the major concerns4142.…”

“…There is accumulating evidence which indicates that ER stress is involved in organ-specific inflammation [100,101], in autoimmunity [93,102] ischemic renal tubular injury [97], and nephrotoxicity of calcineurin inhibitors [97].…”

“…This response can last a long time and causes irreversible tissue damage [8][9][10]. Many drugs, which can block the oxidative stress and inflammatory response cascade, have been used to prevent or decrease I/R injury of the kidney in experimental studies [14,15,[33][34][35][36][37].…”
