Colitis is a growing gastrointestinal inflammation condition that affect individual all around the globe. No specific treatment for colitis condition is available at the moment and this disease can develop into colorectal cancer. Phytochemical in Apium graveolens (AG) are potential as anti-inflammation and antioxidant. This study aimed to investigate the potency of AG ethanolic-extract (EES) in improving acetic acid (AA) – induced colitis in rats. Rats with colitis induced by acetic acid were treated with 5-ASA and several concentrations (100, 300, and 900 mg/kg) of EES. Colitis severity was determined by scoring of macroscopic condition including body weight, colon, and faeces condition. In addition, colon tissues conditions were observed with haematoxylin-eosin staining. Colon permeability is determined by measuring urine glucose level. The observation of the change in Enterobacteriaceae population is investigated using PCR. Apium graveolens ethanolic-extract shows highest improvement of colitis condition at concentration of 900 mg/kg, based on macroscopic observation and disease severity index. At this dose, no blood existed on the faeces and the faeces consistency is solid. Group receiving lower dose of EES still shows blood existence and less solid faeces. The EES dose dependently return the population of Enterobacteriaceae which increase upon AA treatment. Apium graveolens is potential to be use as preventive or treatment for colitis.

Background: Scrophularia striata Boiss. ( S. striata ) is a flowering plant with several therapeutic properties including antiinflammatory, antioxidant, antimicrobial, and wound-healing activity. Regarding the side effects of drugs conventionally used for inflammatory bowel disease (IBD) treatment, we investigated the anticolitis properties of aqueous (SSAE) and hydroalcoholic (SSHE) extracts of S. striata on experimental colitis. Materials and Methods: The colitis was induced using acetic acid (3%) and 2 h before ulcer induction, each group of rats received orally three doses (150, 300, and 600 mg/kg, p.o.) of SSAE or SSHE for the next 5 days. Dexamethasone (1 mg/kg, i.p.) and mesalazine (100 mg/kg, p.o.) were used as reference drugs. Different parameters including weight of colon/height, ulcer index, total colitis index, levels of myeloperoxidase (MPO) and malondialdehyde (MDA) were investigated. Results: Total phenolic contents were 4.3 ± 0.2 and 7.1 ± 0.4 mg/g equivalent to gallic acid for SSAE and SSHE respectively. Three applied doses of SSHE and the highest dose of SSAE (600 mg/kg) could reduce all the macroscopic and pathologic indices of colitis and the levels of MPO and MDA. Two lesser doses of SSAE (150, 300 mg/kg) however, couldn’t diminish the histopathologic features of colitis and the values of MPO and MDA. Conclusions: S. striata , especially SSHE, which also contained more phenolic compounds, had an ameliorating effect on ulcerative colitis and possibly exerts this effect through its antioxidant, antiinflammatory and wound healing properties. Further investigations are required to introduce this plant as a novel alternative herbal drug for colitis treatment.

The goal of this study was to find out how giving whole barley grain to rats that had been given streptozotocin (STZ) affected lipid peroxidation (LPO), the activities of antioxidant enzymes in erythrocytes, and the production of insulin genes in the pancreas. A single shot of STZ (60 mg/kg, i.p.) caused diabetes in the experiment. The oxidative stress was measured by the amount of LPO in the tissue, the amount of reduced glutathione (GSH), and the activities of SOD, CAT, GPx, and GR enzymes in erythrocytes. The most important things that were seen in diabetic control rats were higher blood sugar and LPO levels, lower GSH levels, and lower enzyme functions. When diabetic rats (group G2) were given barley seed whole grain by mouth for 30 days, their LPO level dropped significantly compared to rats that were given STZ (group G3). Furthermore, diabetic rats (group G4) that were fed barley had higher levels of both enzyme- and non-enzymatic antioxidants, as well as higher levels of insulin and regeneration (PRSS1) genes, compared to diabetic normal rats (group G2). The results make it clear that oxidative stress plays a part in causing diabetes and suggest that adding barley to an animal's diet might help protect them.
