Protective and Therapeutic Effects of an IL-15:IL-15Rα-Secreting Cell-Based Cancer Vaccine Using a Baculovirus System

Abstract: This study reports the use of the BacMam system to deliver and express self-assembling IL-15 and IL-15Rα genes to murine B16F10 melanoma and CT26 colon cancer cells. BacMam-based IL-15 and IL-15Rα were well-expressed and assembled to form the biologically functional IL-15:IL-15Rα complex. Immunization with this IL-15:IL-15Rα cancer vaccine delayed tumor growth in mice by inducing effector memory CD4+ and CD8+ cells and effector NK cells which are tumor-infiltrating. It caused strong antitumor immune responses … Show more

“…Nevertheless, IL-15 was shown to promote NK-mediated antitumor response [ 25 , 26 ]. Several preclinical models and clinical trials demonstrated the ability of IL-15 to counteract HCC-induced NK dysfunction, and these findings make it a promising cytokine for cancer immunotherapy [ 27 , 28 , 29 , 30 , 31 , 32 ]. The protective, antineoplastic role of this interleukin was confirmed in our study, as, although it was detected at very low levels in both biological compartments, it was present at higher levels in patients without HCC compared to HCC patients.…”

“…Additional strategies, still in preclinical development, include an autologous cell-based cancer vaccine secreting IL-15 in complex with IL-15Rα. Mice receiving inactivated cancer cells secreting IL-15:IL-15Rα had significant tumor control in both prophylactic and therapeutic cancer models ( 95 ), highlighting the promising potential of IL-15 to improve the anti-tumor response elicited by cancer vaccines.…”
