Protection against cerebral malaria by the low-molecular-weight thiol pantethine

Abstract: We report that administration of the low-molecular-weight thiol pantethine prevented the cerebral syndrome in Plasmodium berghei ANKA-infected mice. The protection was associated with an impairment of the host response to the infection, with in particular a decrease of circulating microparticles and preservation of the blood-brain barrier integrity. Parasite development was unaffected. Pantethine modulated one of the early steps of the inflammation-coagulation cascade, i.e., the transbilayer translocation of p… Show more

“…Other inhibitors such as ticlopidine, pantethine, cystamine, LMP-20, or ROCK inhibitors have also been shown to lower MP levels after treatment (Shouzu et al, 2004;Wassmer et al, 2005;Penet et al, 2008;Antonyak et al, 2012). Although some of these drugs are effective in reducing MP production in patients, they were not capable of bringing down MP levels similar to the healthy control involved in the same study (Nomura et al, 2005(Nomura et al, , 2007.…”

“…Although current pharmacological inhibitors deal with MDR cost effectively, there exist problems of specificity, toxicity and differential modulation 26 with current strategies. MP inhibitors would provide an alternative treatment strategy 48,49 with the added benefit of also targeting other MP associated complications observed in cancer clinically. In this regard, these findings have potential to translate into fundamental outcomes in the science and practice of clinical oncology.…”
