Prostaglandin F2α Induces Goat Corpus Luteum Regression via Endoplasmic Reticulum Stress and Autophagy

Abstract: Corpus luteum (CL) is a transient endocrine tissue that produces progesterone for maintaining pregnancy in mammals. In addition, the regression of CL is necessary for the initiation of the estrous cycle. Extensive research has shown that the prostaglandin F2α (PGF2α) induces the regression of CL in ruminants. However, the mechanisms of endoplasmic reticulum (ER) stress and autophagy in the regression of goat CL induced by PGF2α are still unclear. In this study, ovaries of dioestrus goats and goats that were 3 … Show more

“…Previous studies reported that LC3B protein increased in late-stage bovine corpora lutea (Aboelenain et al, 2015), suggesting that autophagy occurs during induced as well as physiological luteal regression. Similar results have been demonstrated in the corpus luteum of rat and goat (Choi et al, 2011;Grzesiak et al, 2018;Wen et al, 2020). Although autophagy is considered a pro-survival process; we demonstrated that autophagy-related signaling is upregulated during luteolysis, a process that is accompanied by cellular death (Song et al, 2018;Denton and Kumar 2019).…”

“…In response to progesterone, endometrial stromal fibroblasts acquire epithelioid characteristics, such as a highly developed endoplasmic reticulum required for greater secretory function, substantial lysosomes, abundant glycogen and lipids droplets in their cytoplasm 63 . In animal models, the expression of endoplasmic reticulum stress-related proteins and the autophagy marker LC3-II are increased in luteal cells treated with PGF2α 64 . Physiologically, elevated PGF2α in menstruation induces endoplasmic reticulum stress.…”

“…In the ovary, autophagy has been associated with follicle development and atresia as well as luteal formation and regression [ 174 , 176 , 214 , 215 , 216 , 217 , 218 , 219 ]. Recently, Tang et al (2020) found that HIF1α enhances autophagy in the rat luteinized granulosa cells [ 217 ].…”
