The presence and role of prostaglandin moieties in aquatic fauna have been a subject of study for only the last 15 yr. This review discusses the occurrence and physiological functions of these autocoids in a broad range of marine and freshwater fauna. The role of prostaglandins of the E and A series, in particular, in osmoregulation and mineral transport has been studied in several marine invertebrate phyla demonstrating analogy to the roles of these compounds in renal hemodynamics, diuresis, and electrolyte balance in higher terrestrial vertebrates. Prostaglandins of the F series have been implicated in the spawning and hatching processes in a number of marine invertebrates. These findings are discussed in parallel to the body of evidence impl~cating prostaglandins of the E and F series in the reproductive physiology and spawning behavior of a number of teleosts studied. Many of the advances leading to the present state of knowledge of eicosanoid biology in marine and freshwater fauna stem from advances in analytical methodology for the quantitative and qualitative study of these bioactive compounds. Despite these advances, progress in the field of eicosanoid biology in aquatic fauna has proceeded at a slow pace compared to that In terrestrial fauna, leaving significant gaps In current understanding of the roles of these compounds in numerous aquatic species. Continued research in this fleld holds promise for the marine biologist and biochemist, as well as the maricultur~st, if ongoing attempts to monltor and control the cultivation and reproduction of certain aquatic species is to be successful in the future.

The synthesis of prostaglandins (PGs) by several tissue components of brook trout (Salvelinus fontinalis) ovaries was investigated using radiolabeled precursor incorporation and radioimmunoassays for PGE and PGF. PGE2 and PGF2α were synthesized from 14C arachidonic acid (AA) by the follicle walls of mature oocytes. Synthesis was significantly greater in follicles taken postovulation as compared to those sampled prior to germinal vesicle breakdown. However, the most extensive synthesis of PGs occurred with tissue outside of the mature follicles (i.e., extrafollicular[EF]). When incubated with AA, the predominant primary PG produced by this tissue was PGE2. EF tissue could be separated microscopically into two major fractions—immature follicles and stromal tissue. When these tissue fractions were incubated separately with AA the stromal fraction retained the ability to produce PGE2, while the immature follicles increased synthesis of PGF2α. 14C‐Eicosatrienoic acid was converted to PGE1 and PGF1α by EF tissue but in significantly reduced quantities relative to AA. Fatty acid analysis indicated that there were significant quantities of arachidonic, eicosapentaenoic, and docosahexaenoic acids in both follicles and EF tissue, but there was not a significant difference between the two tissues in the absolute amount of arachidonate/mg tissue. In experiments using radioimmunoassays (RIAs), the highest quantities of PGE were measured in incubates containing EF tissue. This supported the results obtained through arachidonic acid incorporation and suggested that this tissue is the primary source of PGE in the brook trout ovary. In contrast, the mature follicules that were undergoing ovulation in vitro produced significant quantities of PGF, whereas the EF tissue production of PGF, measured by RIA, was very low. The results clearly indicate that there are several potential sites for PG synthesis within the trout ovary that differ in their capacity to produce E and F prostaglandins.

This study investigates the regulation of prostaglandin (PG) synthesis in the ovaries of sexually-mature zebrafish (Danio rerio). We examined the ovarian expression of genes within the arachidonic acid (AA) pathway, and the ovarian levels of 17alpha,20beta-dihydroxy-4-pregnen-3-one (17alpha,20beta-P), 17beta-estradiol (E(2)), and PGF(2alpha) in spawning and nonspawning fish during the ovulatory cycle. Real-time RT-PCR analysis revealed that the expression levels of cytosolic phospholipase A(2) (cpla2) and cyclooxygenases (COX)-2 (ptgs2) in ovarian fragments and in isolated full-grown follicles of spawning fish were highest at 6:00 when ovulation was expected to occur. In nonspawning fish, cpla2 expression levels declined over time while ptgs2 expression displayed the same temporal pattern as in spawning fish. Elevated levels of 17alpha,20beta-P in the spawning fish occurred at 3:30, but there were no changes in the nonspawning fish. In other studies conducted to investigate the hormonal regulation of AA pathway genes, fish exposed via the water for 24 or 96 hr to 17alpha,20beta-P or E(2) exhibited reduced ovarian expression levels of COX-1 (ptgs1) and PG E synthase-2 (ptgsl), and E(2) reduced the expression of cpla2. Injection of human chorionic gonadotropin (hCG) (100 IU) led to increased expression levels of cpla2 and ptgs2 at 2 and 18 hr post-treatment, but consistently reduced ptgs1 and ptgsl expression. In these fish, ovarian levels of 17alpha,20beta-P were elevated at all time points and PGF(2alpha) levels in the hCG-treated group were significantly higher than the control fish at 18 hr. Collectively, these in vivo results suggest that gonadotropins and steroids are involved in the regulation of the AA pathway in ovarian follicles of zebrafish.
