Prostaglandin E2 inhibits neutrophil extracellular trap formation through production of cyclic AMP

Abstract: BACKGROUND AND PURPOSEUpon stimulation, neutrophils release their nuclear contents called neutrophil extracellular traps (NETs), which contain unfolded chromatin and lysosomal enzymes. NETs have been demonstrated to play a critical role in host defence, although the role of PGE 2 , a bioactive substance generated in inflammatory tissues, in the formation of NETs remains unclear. EXPERIMENTAL APPROACHThe effects of PGE 2 , agonists and antagonists of its receptors, and modulators of the cAMP-PKA pathway on the … Show more

“…NET formation also occurs during various chronic inflammatory diseases (Kolaczkowska and Kubes, 2013). PGE 2 inhibited NET formation in isolated neutrophils induced by PMA or rapamycin through EP 2 /EP 4 -cAMP receptor signalling (Domingo-Gonzalez et al, 2016;Shishikura et al, 2016), although the extent to which this mechanism operates in vivo remains unknown.…”

“…Small molecule inhibitors of peptidylarginine deiminase inhibit NET generation and have been protective in murine models of lupus and atherosclerosis . Prostaglandin E2 was recently shown to inhibit NET generation . Targeting NETs or specific NET constituents merits further investigation as adjunctive therapies for cancer, initially in tumor‐bearing mice.…”

“…Similar to other cell types, cAMP acts as a key second messenger in the immune system, and in fact, the targeting of the cAMP pathway is widely exploited by the nervous and endocrine systems to modulate the immune response [32][33][34]. cAMP has an overall inhibitory effect on both innate and adaptive immune cell types, including neutrophils [35], macrophages [36], and T and B lymphocytes [37,38]. Prolonged elevation of the intracellular cAMP concentration leads to inhibition of proinflammatory cytokine production [39], macrophage phagocytosis [40], NK cell-mediated cytotoxicity [41], and neutrophil chemotaxis [42].…”
