Prostaglandin D2inhibits fibroblast migration

Abstract: Fibroblasts play an important role in the repair and remodelling processes following injury. Prostaglandin D 2 (PGD 2 ) is a potent mediator in inflammatory processes. In this study, the effect of the PGD 2 on human foetal lung fibroblasts (HFL-1) chemotaxis induced by human plasma fibronectin (HFn) was investigated using the blindwell chamber technique.PGD 2 inhibited HFL-1 chemotaxis to HFn (20 mg?mL -1 ) by 20.8¡3.8% (pv0.05). Checkerboard analysis of HFn-directed migration confirmed that PGD 2 inhibited bot… Show more

“…7). This is interesting as PGD 2 is rapidly converted to 15d-PGJ 2 , which is reported to have anti-inflammatory actions and which inhibits fibroblast migration, whereas PGF 2␣ functions to promote neutrophil chemotaxis and thus has proinflammatory effects (2,17,20,31). It thus appears that as well as COX-2, silica must be acting to induce some of the specific prostaglandin synthases but not others.…”

“…There is little information on the role of PGD 2 or PGF 2␣ on airway remodeling. PGD 2 acts via DP1 and 2 receptors, both of which are present on ASM and fibroblasts (14,32), whereas PGF 2␣ acts on the FP receptor, which has been detected in lung fibroblasts and has been shown to facilitate bleomycin-induced pulmonary fibrosis (47). Because PGF 2␣ did not alter the deposition of ECM proteins, it is possible that it may not be involved in the regulation of these ECM proteins.…”

“…Certain prostaglandins, such as prostacyclin and related analogs prostaglandin (PG) E 2 and PG D 2 , inhibit pulmonary fibroblast migration, proliferation, and collagen synthesis (Keerthisingam et al, 2001;Kohyama et al, 2002;Vancheri et al, 2004). Those agents all activate receptors coupled to G s , implying that cAMP is a negative regulator of fibroblast function.…”
