Prospective Association of Serum and Dietary Magnesium with Colorectal Cancer Incidence

Abstract: Background: Laboratory and epidemiologic research suggests a protective role of magnesium in colorectal cancer development. We estimated the associations of serum and dietary magnesium with colorectal cancer incidence in the Atherosclerosis Risk in Communities (ARIC) study.

“…Second, plasma magnesium concentrations were only assessed at baseline; not only more frequent measurements would have been more informative but would have allowed verifying other potential confounders. Third, although previous studies suggested that the association between magnesium and risk of cancer varied between different cancer types, including colorectal [18], prostate [19, 24], and lung [25] cancers, there was no significant heterogeneity observed across different cancer subtypes in the current study. However, our current study was underpowered to investigate individual cancers.…”

“…Research findings to date on the association between magnesium status and incident risk of cancer remain inconsistent. Polter et al [18] found a higher risk of colorectal cancer with lower serum magnesium among females but not males. Dai et al [24] suggested low serum magnesium levels was significantly associated with high-grade prostate cancer.…”

“…Mg deficiency, due to either low Mg intake or excessive Mg waste, can be responsible of defects in DNA repairing mechanisms leading to genomic instability [ 11 ]. Hypomagnesaemia is frequent in patients with CRC [ 12 , 13 ] and serum Mg level detection was proposed as a biomarker of efficacy and outcome in patients with advanced CRC treated with cetuximab [ 14 , 15 ] and bevacizumab [ 16 ]. Hypomagnesaemia in cancer patients might be due to iatrogenic causes [ 17 ] or to the tumour itself, which may act as a Mg trap [ 18 ].…”
