Proposal and application of an ecotoxicity biotest based on escherichia coli

Abstract: The authors propose the use of an ecotoxicity biotest based on Escherichia coli for the sanitary control of water. The sensitivity of the proposed model was evaluated using a group of representative toxicants, and its response (synergism, additive or antagonism) to toxicants was observed. Finally, the biotest was applied to 124 water samples from rivers, underground sources, drinking water and wastewater. The results of the biotest were compared with the sanitary classification of the waters.

“…39,40 The toxic effect is assessed by testing growth inhibition of the bacterial strain Escherichia coli K12 W3110 thy − (CECT 416, Spanish Type Culture Collection). The assay was carried out in tubes containing 10 ml of Vogel-Bonner culture medium supplemented with 50 mg 1 −1 of thymine and 20 g 1 −1 of glucose.…”

“…Some data regarding the predictive power of this biotest have been published previously. 39,40 The objective of the present study was to assess the toxicity of a group of newly synthesized DTCs using the IGEC bioassay, and to evaluate statistical models based on quantitative structure-activity relationships for estimating the possible relationship between some chemical characteristics of these compounds and their degree of toxicity.…”

“…As can be seen in Table I, subinhibiting concentrations of nickel and methyl-parathion were used with a view to the possible application of the measurement of the E. coli peroxidation levels as a marker for ecotoxicity biotests. The sensitivity of this method would ideally be equal to or greater than that of simpler techniques, such as those based on the inhibition of growth (Espigares et al, 1990).…”

“…The bacterial strain Escherichia coli K-12 W3110 thy-F-was incubated in Vogel-Bonner medium supplemented with thymine (50 mg/L), glucose (20 g/L), and peptone (0.5 g/L) (Davies et al, 1980;Maron and Ames, 1983). Toxicants were added in previously determined subinhibiting concentrations of 40 pg/mL nickel and 62.5 pg/mL methyl-parathion (Espigares et al, 1990;Odberg-Ferragut et al, 1991).…”
