Propofol directly induces caspase-1-dependent macrophage pyroptosis through the NLRP3-ASC inflammasome

Abstract: Propofol infusion syndrome (PRIS) is an uncommon life-threatening complication observed most often in patients receiving high-dose propofol. High-dose propofol treatment with a prolonged duration can damage the immune system. However, the associated molecular mechanisms remain unclear. An increasing number of clinical and experimental observations have demonstrated that tissue-resident macrophages play a critical role in immune regulation during anaesthesia and procedural sedation. Since the inflammatory respo… Show more

“…Reactive oxygen species (ROS) such as hydrogen peroxide and hydroxyl ions are created through the reduction of oxygen during molecular processes. ROS play an important role in both pyroptosis and necroptosis [ 54 , 55 , 56 , 57 , 58 , 59 , 60 ]. In pyroptosis, ROS commonly serve as the second signal leading to inflammasome activation, which forms as a precursor to caspase-1 activation, Gasdermin D cleavage, and subsequent membrane rupture.…”

“…NLRP3 inflammasomes are an activating platform of caspases, consisting of NLRP3, ASC and caspase-1 (13). The activation of NLRP3 inflammasomes is closely associated with tumor formation (28).…”

“…NLR family pyrin domain-containing 3 (NLRP3) inflammasomes are multi-protein complexes composed of the intrinsic intracellular immune receptor NLRP3, the adaptor protein apoptosis-associated speck-like protein containing a CARD (ASC) and protease caspase-1 (13). The assembly of this complex can induce the maturation and secretion of pro-inflammatory factors, such as IL-1β and IL-18, thus promoting the development of an inflammatory response (14).…”

“…In most studies, various types of programmed cell death are considered independent, discrete, and unrelated. Apoptosis and pyroptosis are different in morphology, function, and mechanism [40]. On a cellular level, while membrane shrinking and the formation of vesicular bodies (apoptotic bodies) is the outcome of apoptosis.…”
