Properties of Parallel Tetramolecular G-Quadruplex Carrying N-Acetylgalactosamine as Potential Enhancer for Oligonucleotide Delivery to Hepatocytes

Abstract: The development of oligonucleotide conjugates for in vivo targeting is one of the most exciting areas for oligonucleotide therapeutics. A major breakthrough in this field was the development of multifunctional GalNAc-oligonucleotides with high affinity to asialoglycoprotein receptors (ASGPR) that directed therapeutic oligonucleotides to hepatocytes. In the present study, we explore the use of G-rich sequences functionalized with one unit of GalNAc at the 3′-end for the formation of tetrameric GalNAc nanostruct… Show more

“…Murine primary hepatocytes were transduced with all vectors at a MOI of 1E6, leveraging the targeting capability of GalNAc sugar derivatives to bind to the ASGPR, which is abundantly expressed on hepatocytes. [33] Figure 3 illustrates that transgene (i.e. GFP) expression efficiency 48h after transduction, by FACS analyses.…”

“…In the original article [ 1 ], there was a mistake in the position of one of the OH of N-acetylgalactosamine. In the original article [ 1 ], the OH was in an equatorial position and it should be in an axial position. This mistake affected Scheme 1 and Scheme 2.…”
