Properties of a Proteus morganii Bacteriocin

Abstract: SUMMARYThe bacteriocin produced by Proteus morganii strain MR 336 is a thermolabile glycoprotein with a sedimentation constant of 4 0 . Activity is dependent on the integrity of the complex and the bacteriocin is inactivated by oxidation. Bacteriocinogeny could not be transmitted to other strains of P. morgunii but was eliminated by 30,ug./ml. acridine orange at pH 7.45. Bacteriocin-resistant mutants of a sensitive P. morganii strain were readily obtained.

“…Bactericidal effect of Bac R. on a group A streptococcal strain. The bactericidal effect of Bac R1 against Streptococcus pyogenes strain M57 was monitored during growth of the streptococci in HI broth containing 20 AU of bacteriocin per ml. As a control, phosphate buffer was added to an identically treated culture.…”

“…Staphylococcins can be sensitive to mechanical agitation (11), but 30 min of continuous high-speed vortexing failed to reduce the activity of a Bac R. solution. Other bacteriocins are inactivated by oxidation (20), but Bac R. was not inactivated after incubation at 370C in a final concentration of 10 mM H202 for 2 h. Several enzymes were then interacted with Bac R1 to determine whether they affected bactericidal activity. Lysostaphin did not reduce Bac R. activity.…”

“…Although the action of acnecin seems to be different in this respect, it has a notable feature in common with other bacteriocins. As Jetten and Vogels pointed out (14), there are many similarities in amino acid composition among bacteriocins from different species of S. aureus (14), S. marcescens (8), Lactobacillus fermenti (5), and Proteus morganii (30) and colicin (11). The most striking similarity is high content of aspartic acid, glutamic acid, glycine, and alanine.…”

“…The protracted accumulation of morganocin in Proteus cultures reflected its stability in the media, in contrast to the bacteriocin produced by P. morganii MR336, which had to be stabilized by addition of sodium sulfite (27). A long lag period after induction (maximum production 36 to 48 h after induction) has also been described for streptococcin A (28).…”

“…Many are coded for by plasmids which have been identified by isolation (25), elimination (16,27), or mobilization as a result of their sex factor activity (22) or that of a mobilizing plasmid (29). Several bacteriocins have been purified and characterized to varying degrees, and their mode of action has been established (see 9).…”
