Propagation and Maintenance of Cancer Stem Cells: A Major Influence of the Long Non-Coding RNA H19

Abstract: Cancer stem cells (CSCs) represent a rare population of tumor cells that exhibit stem cell properties with the abilities of self-renewal and differentiation. These cells are now widely accepted to be responsible for tumor initiation, development, resistance to conventional therapies, and recurrence. Thus, a better understanding of the molecular mechanisms involved in the control of CSCs is essential to improve patient management in terms of diagnostics and therapies. CSCs are regulated by signals of the tumor … Show more

“…Long non-coding RNAs (lncRNAs) are a heterogeneous class of transcripts longer than 200 nucleotides with limited protein-coding potential [158,159]. CAFs were shown to transfer lncRNA H19 to neighboring colorectal cancer cells [151,[160][161][162] and promote the stemness via activating Wnt/β-catenin signaling causing resistance to oxaliplatin-mediated apoptosis [163]. Exosomal lncRNA enhance invasion, migration, proliferation and inhibit apoptosis in cervical cancer cells and in NSCLC [164,165].…”

“…Furthermore, in colorectal cancer, the lncTCF7 interacts with subunits of the SWI/SNF chromatin remodeling complex, regulating the transcription of the TCF7 gene and activating the Wnt cascade, involved in stem cell self-renewal ( 229 ). H19 is another lncRNA overexpressed in many cancers and confer stem-like properties in correlation with stem cell markers like SOX2, OCT4, NOTCH1, c-Myc, and ABCG2 ( 230 232 ).…”

“…They have both multicellular differentiation potential and self-renewal ability. CSCs reprogram relevant cells in the tumor microenvironment to favor tumor development while suppressing the tumor-killing effect of immune cells (109,110). Chen et al found that targeting CMTM6 suppresses stem cell-like properties in HNSCC (18).…”
