One method to overcome the difficulties involved in reproducing first generation Senegalese sole that are born in captivity (F1) is to obtain gametes by stripping and then using them for artificial fertilization. The aim of this paper was to determine an appropriate protocol for hormonal induction of ovulation and artificial fertilization. To induce ovulation, two different GnRHa administration methods, weekly injections versus a single GnRHa implant, were compared. The number of spawns per female and the relative fecundity were significantly higher in females that received the hormonal therapy through repeated injection (6.3 ± 0.8 spawns per female and 574.9 ± 67.2 x 10 3 eggs kg -1 BW) than those treated with a single implant (2.7 ± 0.6 spawns per female and 134.6 ± 40.9 x 10 3 eggs kg -1 BW). The lapsed time to ovulation and the changes in fertilization and hatching rates of eggs that were retained in the ovarian cavity after ovulation were determined after GnRHa treatment. At 16°C, ovulation was detected at 41-44 h post-treatment in 70% of the females. The highest fertilization and hatching rates were obtained during the first 3 h after ovulation, and A C C E P T E D M A N U S C R I P T ACCEPTED MANUSCRIPT2 they significantly decreased thereafter. The results suggested that the GnRHa injection was the most appropriate administration method for the induction of ovulation. Ovulated eggs inside the female were viable for a short time, and therefore it is important to determine the time of ovulation after hormonal treatment.

Gonadotropin-releasing hormone in Cyprinidae as in other Vertebrates functions as a brain signal which stimulates the secretion of luteinizing hormone from the pituitary gland. Two forms of gonadotropin-releasing hormone have been identified in cyprinids, chicken gonadotropin-releasing hormone II and salmon gonadotropinreleasing hormone. Hypohysiotropic functions are fulfilled mainly by salmon gonadotropin-releasing hormone. The only known factor having an inhibitory effect on LH secretion in the family Cyprinidae is dopamine. Most cyprinids reared under controlled conditions exhibit signs of reproductive dysfunction, which is manifested in the failure to undergo final oocyte maturation and ovulation. In captivity a disruption of endogenous gonadotropinreleasing hormone stimulation occurs and sequentially that of luteinizing hormone, which is indispensible for the final phases of gametogenesis. In addition to methods based on the application of exogenous gonadotropins, the usage of a method functioning on the basis of hypothalamic control of final oocyte maturation and ovulation has become popular recently. The replacement of natural gonadotropin-releasing hormones with chemically synthesized gonadotropin-releasing hormone analogues characterized by amino acid substitutions at positions sensitive to enzymatic degradation has resulted in a centuple increase in the effectiveness of luteinizing hormone secretion induction. Various induction protocols were compared with the aim to reach ovulation in northern pike (Esox lucius L.). The effectiveness of salmon D-Arg 6 Pro 9 NEt-GnRHa at two doses in combination with various additives were tested: 50 and 100 µg kg -1 (groups GnRHa 1 and 2), GnRHa at 50 and 100 µg kg -1 in combination with 8 mg kg -1 metoclopramide (groups 3 and 4), GnRHa at 50 and 100 µg kg -1 emulsified in Freund´s incomplete adjuvant (FIA) (groups 5 and 6), and GnRHa at 50 and 100 µg kg-1 emulsified with FIA 8 mg kg -1 and metoclopramide (groups GnRHa 7 and 8). These treatments were compared to indoor controlled conditions (Group ICC, injected with saline solution, without hormonal treatment) and two traditional treatments: 3 mg kg -1 carp pituitary (Group CP) and ambient outdoor conditions (Group AOC, injected with saline solution only, without hormonal treatment).All fish in Group CP and 70% of those in Group AOC ovulated. The latency periods for Group CP was 96±4 h and 264±58 h for Group AOC. One Group-3 female ovulated spontaneously 108 h post-injection, whereas none of the other GnRHa injected or Group ICC fish ovulated. The pseudogonadosomatic index was 19.3±5.9%; 18.5% and 17.8±7.5% in the AOC, Group 3 and CP, respectively. The fertilization rate reached 88.6±4.5%; 85.5±12.0% and 66.0±13.7% in AOC, Group 3 and CP, respectively. The hatching rate was 68.6±9.9%; 65.5±7.5% and 54.4±8.0% in AOC, Group 3 and CP, respectively.The ovarian fluid pH was significantly higher (8.27±0.03) in AOC compared to CP group (8.11±0.02) (P>0.05). Ovarian fluid of the spontaneously ovulated Group-3 fish had a pH of 8.35. There were no significant differences in egg size and weight among successfully reproduced groups -AOC and CP (P>0.05).
