Promoter Methylation Controls the Expression of MAGE2, 3 and 4 Genes in Human Cutaneous Melanoma

Abstract: Cancer-testis antigens expressed by different-histotype transformed cells are suitable targets for tumor immunotherapy. However, their heterogeneous expression in neoplastic lesions limits the eligibility of patients for cancer-testis antigen-directed vaccination, and low levels of cancer-testis antigens' expression may impair immune recognition of malignant cells. Because of the primary clinical relevance of cancer-testis antigens' expression in neoplastic tissues, 68 unrelated or sequential metastatic lesion… Show more

“…To understand the activation of MAGED4 in glioma cells is related to the DNA methylation, glioma cells (SHG44 and A172) were treated with 5-AZA-CdR, a DNA demethylating agent which has been use to upregulated to the expression of genes silenced by the hypermethylation of their promoter (Weber et al, 1994;Sigalotti et al, 2002). Our result showed the reaction to 5-AZA-CdR in the two cells were various.…”

“…The X-linked MAGEA3 gene is hypomethylated and overexpressed in melanoma (Sigalotti et al, 2002), and the methylation status of the regulatory regions of SH2D1A, the X-linked lymphoproliferative disease gene, correlates with tissue-specific expression (Parolini et al, 2003). Hypomethylation of the CpG islands associated with the MAGEA3 and SH2D1A genes also correlated with increased expression, 2.54-and 3.05-fold, respectively (Figures 4b, d and 3).…”

“…Samples were scored 7, no RT -PCR product detectable; +, expression level 510% to that of the appropriate reference cell line; ++, expression level 410% to that of the appropriate reference cell line. Competitive PCR for MAGE-3 and b-actin was performed as described (Sigalotti et al, 2002).…”

“…Furthermore, due to the demonstrated role of DNA methylation in regulating gene expression (Jones and Takai, 2001), and since promoter methylation is involved in the expression of MAGE genes in human melanoma (De Smet et al, 1996, 1999Sigalotti et al, 2002), we investigated the role of the DNA hypomethylating agent 5-aza-2'-deoxycytidine (5-AZA-CdR) in regulating the differential distribution of CTA and their constitutive levels of expression in MM.…”
