Proline and hydroxyproline deposited on silver nanoparticles. A Raman, SERS and theoretical study

Abstract: The Raman and surface‐enhanced Raman scattering (SERS) spectra of l‐proline (Pro) and trans‐4‐hydroxy‐ l‐proline (Hyp) were recorded. SERS spectra were obtained on colloidal Ag prepared by reduction with hydroxylamine. Allowing sufficient time for Pro and Hyp to adjust in the colloidal solution resulted fundamentally in obtaining unique and reproducible SERS spectra. Hyp stabilizes on the surface more rapidly than Pro. The spectral analysis indicates that Pro interacts with the Ag surface through the carboxyla… Show more

“…The influence of the Pro residue oscillations on the BK SERS spectra could not be excluded because the ~1467 [amide IIp marker of relatively strong prolyl C=O interactions], ~1443 [amide IIp marker of non‐hydrogen‐bonded C=O acceptor], ~1255 [amide III], ~874 [ρ r (CH 2 ) ring ], ~745 [ρ r (CH 2 ) ring ], and ~679 cm −1 [amide V]) spectral features are characteristic of the tertiary amide bond and Pro ring . The 935 (which was rather weak in the Ag and Au spectra) and 1378 cm −1 bands are due to the C ‐terminal deprotonated carboxylate group.…”

“…The Hyperchem program was used . The combination of EHT with molecular mechanics was able to give, for example, a good qualitative explanation of our previous SERS works in peptides, nanotubes, tryptophan, lysine, humic acids and 4‐hydroxyproline and proline interacting with Ag surfaces.…”
