“…Using liquid scintillation counting methods, Horwitz, Stastny, and Ziff suggested that increased mononuclear uptake of tritiated thymidine (Tdr-8H)1 occurred especially in the acute phase of an inflammatory disease and may reflect either a response to nonspecific tissue injury, or antigenic stimulation (2). Radioautographic studies were in agreement with other investigators who have shown that DNA-synthesizing cells are large mononuclear cells (3,4) which have been called "atypical lymphocytes" (5).…”

“…Many of these were morphologically similar to the "stress lymphocytes" of Frank and Dougherty (10), and as a group to the "atypical lymphocytes" of Wood and Frenkel (5). 35% of these large nonphagocytic mononuclears were DNAsynthesizing cells similar to those originally described by Bond and coworkers (3) and recently by Horwitz, Stastny, and Ziff (2). Tdr-2H-labeled large phagocytic cells (monocytes) or small lymphocytes were rare.…”

“…At this time, activity in terms of thymidine uptake was 5-to 10-fold less, and mitoses, labeled or unlabeled were never found before the 32nd hr of culture. It was considered that this initial labeling activity did not represent a specific immune response since control cultures behaved in a similar manner, but rather, probably reflected either an adjustment of the blood leucocytes to the culture conditions, or the known low level of DNA synthesis in cells of normal peripheral blood (22). By the 44th hr after the mixed cell cultures were initiated (group C, 33 ~ hr) the levels of 3HTdR incorporation in them were significantly elevated over those in the control cultures, and the first labeled nuclei (0.4%) and mitotic figures appeared.…”

“…This opinion has received support from many experimental observations including the lack of deoxyribonucleic acid (DNA) synthesis in these cells (1), their low rate of protein synthesis (2,3), and their long life span (4,5). On Received for publication 15 September 1967 and in revised form 15 November 1967. cellular (6) and humoral (7) immune reactions has been firmly established.…”
