Severe acute respiratory syndrome (SARS) emerged from China as an untreatable and rapidly spreading respiratory illness of unknown etiology. Following point source exposure in February 2003, more than a dozen guests infected at a Hong Kong hotel seeded multi-country outbreaks that persisted through the spring of 2003. The World Health Organization responded by invoking traditional public health measures and advanced technologies to control the illness and contain the cause. A novel coronavirus was implicated and its entire genome was sequenced by mid-April 2003. The urgency of responding to this threat focused scientific endeavor and stimulated global collaboration. Through real-time application of accumulating knowledge, the world proved capable of arresting the first pandemic threat of the twenty-first century, despite early respiratory-borne spread and global susceptibility. This review synthesizes lessons learned from this remarkable achievement. These lessons can be applied to re-emergence of SARS or to the next pandemic threat to arise.

Development and Validation of a High-Throughput Screen for Inhibitors of SARS CoV and Its Application in Screening of a 100,000-Compound Library

The authors have developed a high-throughput screen (HTS) that allows for the identification of potential inhibitors of the severe acute respiratory syndrome coronavirus (SARS CoV) from large compound libraries. The luminescent-based assay measures the inhibition of SARS CoV-induced cytopathic effect (CPE) in Vero E6 cells. The assay was validated in 96-well plates in a BSL3 containment facility. The assay is sensitive and robust, with Z values > 0.6, signal to background (S/B) > 16, and signal to noise (S/N) > 3. The assay was further validated with 2 different diversity sets of compounds against the SARS CoV. The "hit" rate for both libraries was approximately 0.01%. The validated HTS assay was then employed to screen a 100,000-compound library against SARS CoV. The hit rate for the library in a single-dose format was determined to be approximately 0.8%. Screening of the 3 libraries resulted in the identification of several novel compounds that effectively inhibited the CPE of SARS CoV in vitro-compounds which will serve as excellent lead candidates for further evaluation. At a 10-µM concentration, 3 compounds with selective indexes (SI 50 ) of > 53 were discovered. (Journal of Biomolecular Screening 2007:33-40)

ÖzetCoronavirüsler (CoVs) çok çeşitlilik gösteren bir virüs ailesidir. CoV'lar konak hücrelerle birçok aşamada etkileşime geçerler ve konak hücrenin değişik mekanizmalarını kullanarak enfeksiyonlarını ve replikasyonlarını gerçekleştirirler. CoV'ların moleküler biyolojisiyle ilgili olarak birçok şey bilinmektedir. Ancak CoV'ların moleküler biyolojisini daha iyi anlamak için daha çok bilgiye ihtiyaç vardır ve CoV'lara karşı geliştirilmiş herhangi etkili bir ilaç tedavisi bulunmamaktadır. CoV'ların moleküler biyolojisini daha iyi anlama adına son yıllarda yapılan araştırmalarda birçok teknik ve yöntem kullanılmıştır. Moleküler biyolojik araştırmalar açısından fare hepatit virüsü en çok çalışılan CoV'lardır ve Coronaviridae ailesi üyelerini araştırmak için iyi bir model sistemdir. Bu derleme CoV'lerin replikasyonunu, transkripsiyonunu ve tekrar bir araya getirilmesini anlatmaktadır. Anahtar kelimeler: Coronavirüs; replikasyon; transkripsiyon; toplanma Abstract Coronaviruses (CoVs) are a diverse family of viruses that interact at multiple levels with components of host cells taking this advantage of some of the cellular machineries for replication and proliferation. A lot is known about the molecular biology of CoVs but more information needs to be learned because no effective treatments against these viruses are available. The challenge now is to incorporate advance techniques in the investigative efforts done to understand further the biology of CoVs. In terms of molecular biology, mouse hepatitis virus (MHV) is the best-studied CoVs, and it is consequently considered a model system for the family of Coronaviridae. This review described replication, transcription, and assembly of CoVs.
