“…Many recent studies have noted the bene®t of seeding preparations of lacrimal acinar cells onto Matrigel-coated culture plates (Kelleher et al, 1991;Meneray et al, 1994;Millar et al, 1996;Chen et al, 1998). Recent studies by the authors group have established procedures for the isolation of highly puri®ed preparations of lacrimal acinar cells (Guo et al, 2000) and methods which enhance the proliferation of the lacrimal acinar cells (Scho Ènthal et al, 2000). Prior to this time lacrimal acinar cells could be established and maintained in culture for short periods of time, approximately 7 days, with minimal proliferation.…”

“…The most recent methods (Scho Ènthal et al, 2000) combine the bene®ts of using Hepato Stim 1 Medium [supplemented with epidermal growth factor (EGF) and fetal bovine serum (FBS)] and Matrigel, a solubilized preparation known to be rich in extracellular matrix molecules derived from the basement membranes of the Engelbreth±Holm±Swarm (EHS) mouse sarcoma line (Kleinman et al, 1982). Unlike past studies in which Matrigel was used to coat the surfaces of culture plates, the authors have devised methods to coat the preparations of puri®ed, suspended, lacrimal acinar cells with Matrigel.…”

“…Previous studies showed that the maintenance of the morphology and secretory function of lacrimal gland cells in vitro critically depends on the use of an extracellular matrix and on the composition of growth factors and nutrients in the culture medium [7,16,20,21,23,26].…”

“…The rate of EGF secretion into the tear fluid rapidly increases in response to stimulation (van Setten, 1990;Yoshino et al, 1997), and lacrimal EGF mRNA expression increases in response to corneal injury (Wilson et al, 1999). EGF stimulates proliferation of lacrimal acinar cells in primary culture (Schönthal et al, 2000). EGF also has been reported to increase protein secretion by acinar cells from rat lacrimal glands (Dartt, 2001;Tepavcevic et al, 2003).…”
