Proliferation of Guinea Pig Tracheal Epithelial Cells Induced by Calcitonin Gene-related Peptide

Abstract: Calcitonin gene-related peptide (CGRP) is contained within and secreted by nerves and neuroepithelial bodies in the airway epithelium. To determine whether CGRP is mitogenic for airway epithelial cells, tracheal epithelial cells isolated from 26 guinea pigs were grown in primary culture for 2 days. Subconfluent cells were exposed to 10(-13) to 10(-9) M CGRP for 4 h and then returned to CGRP-free medium. Proliferation was quantified by direct cell count and by measurement of fractional labeling with the thymidi… Show more

“…tor cells. Interestingly, the mitogenic effect of CGRP on guinea pig tracheal epithelial cells has been shown to be lost after 3 days of exposure (28), supporting the notion that sustained CGRP signals in CF SMGs could alter the phenotype of slow-cycling progenitor cells in the glandular niche.…”

“…CGRPs have been shown to stimulate the proliferation of airway epithelial cells in vitro (27,28). Given this finding, we hypothesized that induction of glandular CGRP might be involved in signaling progenitor cell expansion in SMGs following airway injury and that dysregulation of this mechanism in CF may be responsible for formation of a facultative slow-cycling progenitor cell niche in the SAE.…”

“…In an in vitro model using lung buds, treatments with bombesin resulted in increasing branching morphogenesis (King et al, 1995). CGRP causes proliferation of guinea pig tracheal epithelial cells in primary culture through stimulation of a specific receptor (White et al, 1993). We have identified these peptides in NEBs surrounded by clusters of CCSPimmunolabeled cells.…”

“…CGRP induces degranulation in human neutrophils; promotes neutrophil adherence to endothelial cells (Richter et al 1992;Zimmerman, Anderson, and Granger 1992); and modulates migration and proliferation of cells, including tracheal epithelial cells (White et al 1993;Mikulec and Tanelian 1996;Kim et al 1995).…”
