Proliferation and differentiation of human adipose-derived mesenchymal stem cells (ASCs) into osteoblastic lineage are passage dependent

Abstract: Osteogenic potential of ASCs is impaired by successive passaging and may not serve as a useful clinical source of osteogenic ASCs past P2.

“…The number of passages (up to passage 4) to obtain a pure population of adipose-derived stromal cells (before seeding onto the human acellular collagen matrix) with optimal angiogenic properties (VEGF secretion in hypoxia) cannot be exceeded. Several alternatives of improvement (to obtain passage 4 as quick as possible) can be postulated, such as a larger amount of harvested adipose tissue, improvement of cell expansion by the use of platelet lysate, 42,43 the use of successive subcultivation, 44 and others. Preliminary results (data not shown) demonstrated that the time of the expansion of adipose-derived stromal cells up to passage 4 can be significantly reduced by more than 50 percent.…”

“…Under conventional culture conditions in a dish or flask, cells are plated in 2-dimensional layer, which builts a fairly artificial environment, that has been shown to be detrimental to phenotype preservation in various cell lines. Loss of differentiation potential into osteogenic and adipogenic lineage has been described for adipose-derived stem cells 20,21 during monolayer culture. On the other hand, by 3D-cultivation in microfibers, human pluripotent stem cells have preserved their undifferentiated phenotype to a significantly higher passage than in culture on a matrigel surface 22 .…”

“…Mesenchymal stem cells are one of the most important adult stem cells that have multivariate characteristics, fibroblastic morphology, high proliferation potential, and adhesion properties to the floor of plastic containers, selfrenewal, and the ability to differentiate into multiple cell lines (6)(7)(8)(9)(10). Adiposed-drived mesenchymal stem cells, in addition to the mentioned characteristics, also have other advantages, such as the lack of ability to proliferate and differentiate in long time cultures and the availability of large reserves and easy access to a large amount of fat through sampling and liposuction (11)(12)(13)(14)(15).…”
