Prolactin and Thyrotropin Responses to Thyrotropin-Releasing Hormone in Patients with Secondary Amenorrhea: The Effect of Bromocriptine

Abstract: Prolactin (PRL) and thyrotropin (TSH) responses to a 200 mug intravenous thyrotropin-releasing hormone (TRH) bolus were measured by radioimmunoassay in 11 women with hyperprolactinemic amenorrhea and 9 with normoprolactinemic amenorrhea. In all cases, the tests were carried out under basal conditions and repeated during bromocriptine treatment. In women whose basal PRL level was normal; TRH caused a maximal PRL increment of 85 +/- 25.2 mug/l (mean +/- SE), while those women whose basal PRL level was raised sho… Show more

“…The lack of effect of nomifensine on TRH-induced TSH release is not unexpected, since acute administration of bromocryptine itself is ineffective (Del Pozo et aL, 1972;Del Pozo etaL, 1973;Hirvonen et al, 1975). Both chronic oral administration of L-DOPA and acute DA infusion, however, inhibit the TSH response to TRH in man (Besses et aL, 1975;Noel et al, 1973;Spaulding etal., 1972), suggesting an overall inhibitory dopaminergic control of TSH secretion.…”

“…In vivo data in humans have shown the ability of dopamine or the dopaminergic agent bromocriptine to block PRL secretion induced by different stimulants (15)(16)(17). Whether the same is true concerning the stimulating effect of ghrelin on PRL release has not been studied.…”

“…Αν και η TRH είναι γνωστός διεγέρτης της έκκρισης της PRL στον άνθρωπο (Hirvonen et al, 1976;Andreassen et al, 1979;Kauppila and Ylikorkala, 1982), ένα πεπτίδιο που διεγείρει την έκκριση της PRL αποµονώθηκε πρόσφατα από τον υποθάλαµο βοοειδών (Hinuma et al, 1998). Παρ' όλα αυτά, η συµµετοχή του πεπτιδίου αυτού στο φυσιολογικό µηχανισµό έκκρισης της PRL δεν έχει διευκρινισθεί (Lin, 2008).…”

“…Η µετοκλοπραµίδη είναι γνωστός διεγέρτης της έκκρισης της PRL στον άνθρωπο ανταγωνιζόµενη τη δράση της ντοπαµίνης (Judd et al, 1976;Cunha-Filho et al, 2001). Μέχρι σήµερα, η ντοπαµίνη είναι η µόνη γνωστή υποθαλαµική ουσία, η οποία πιστεύεται ότι ρυθµίζει την έκκριση της PRL από την υπόφυση (del pozo and Brownell, 1979;Reymond and Porter, 1985;Ben-Jonathan and Hnasko, 2001), αν και η TRH είναι επίσης πιθανόν να συµµετέχει (Hirvonen et al, 1976;Andreassen et al, 1979;Kauppila and Ylikorkala, 1982), όπως περιγράφηκε παραπάνω.…”
