Proinflammatory effects and mechanisms of calprotectin on human gingival fibroblasts

Abstract: Our findings suggest that calprotectin exerts proinflammatory effects on HGFs via the S100A9 subunit and TLR4-mediated NF-κB and MAPK signaling pathways.

“…Recent studies have identified S100A9 as a ligand of the receptor for advanced glycation end products (RAGE), which plays a regulatory role in pro-inflammatory responses 25 , 26 . Moreover, S100A9 has been shown to be closely related to the MAPK signaling pathway, which promotes cancer development and tumor spread via inflammatory pathways 8 , 27 ; however, the mechanism underlying the regulatory role of S100A9 in SM progression was unclear.…”

“…In response to TLR4 stimulation, S100A9-deficient neutrophils exhibited impaired production of cytokines ( 75 ). S100A9 and S100A8/A9 significantly upregulated IL-6 and IL-8 expression in human gingival fibroblasts via the S100A9 subunit ( 76 ). (Figure 1 demonstrates the S100A8/A9-induced inflammation signaling pathway involved in TLR4, MyD88, and MAPKs.)…”

“…1 HLA-B 57 was identified as an independent resistance indicator for generalized periodontitis. 3 Apart from this finding, HLA-B57.01-positivity, affecting ∼5 to 11% of the Caucasian population, was identified as major risk factor for development of the abacavir-hypersensitivity reaction. 11,12 Abacavir is a guanosine-analogue, broadly used as a reverse transcriptase inhibitor in antiretroviral combination therapy.…”
