Progression of Prothrombin Induced by Vitamin K Absence-II in Hepatocellular Carcinoma

Abstract: Hepatocellular carcinoma (HCC) is the fifth most common cancer and the third leading cause of cancer-related death worldwide. Due to the lack of efficient tools for early detection, asymptomatic HCC patients are diagnosed at an advanced stage, leading to a poor prognosis. To improve survival, serum biomarker prothrombin induced by vitamin K absence-II (PIVKA-II) was under investigation. PIVKA-II is an abnormal protein produced in HCC. The coagulation function was insufficient due to the lack of Gla residues. E… Show more

“…AFP and PIVKA‐II are significant in the evaluation of treatment efficacy and prognosis of HCC. Patients with elevated PIVKA‐II are more prone to recurrence and metastasis 11,12 . However, there are few reports on the correlation between serum AFP and PIVKA‐II in the prognosis of HCC patients.…”

“… 68 , 292 , 293 Of note, several emerging biomarkers, such as prothrombin induced by vitamin K absence-II (PIVKA-II), featuring a higher sensitivity and specificity than AFP have not yet been widely deployed in preclinical and clinical settings. 294 The limitation of geographic populations and underlying liver disease, and the uncertain biomechanisms to some extent contribute to the existing disappointing situation. Further well-designed, large-scale, and international multicenter prospective clinical studies as well as in-depth mechanistic studies are warranted to validate the clinical power of these biomarkers.…”

“…PIVKA-II in combination with serum α-fetoprotein (AFP) enhanced the accuracy of surveillance of HCC in high-risk populations. Additionally, PIVKA-II acts as a marker for the assessment of response to treatment in HCC ( Yang et al, 2021 ). Hitomi et al investigated the antitumor effects of vitamin K2 on athymic BALB/c-nu/nu mice.…”
