Progress in the Synthesis of 4-Hydroxy-2-pyridone Derivatives

Abstract: 4-Hydroxy-2-pyridones are embedded as common structural units of many natural products, which exhibited good or excellent biological activities such as antifungal, antibacterial, and cytototoxic activity and so on. The class of compounds has stimulated a great deal of interest on synthetic chemists due to their structural novelty, molecular diversity, and promising biological activities. In this paper, the recent progress in the synthesis of 4-hydroxy-2-pyridones has been reviewed.

“…Among the numerous intriguing molecules that we have isolated and identified from P. dives , a particular class of dopamine derivatives featuring a benzothiazole fragment ( 6 and 7 ) has captured our attention. The benzothiazole skeleton, owing to its flexible substituent positions and remarkable bioactivity, is frequently incorporated into the design of pesticide molecules [8–13] . It has also played a significant role in the development of several commercial pesticides, including benzthiazuron ( 8 ), [14] dufulin ( 9 ), [15] mefenacet ( 10 ), [16] fenthiaprop ( 11 ), [17] and methabenzthiazuron ( 12 ) [18] etc (Figure 1B).…”

“…Substituted 2-pyridones posses interesting biological and pharmacological activities. [1,2] For example, 2-pyridones have shown antimicrobial, [3] antidepressant, [4] cardiotonic, [5] and anticancer activities. [6] A few reports have appeared on the synthesis of 2-pyridones, which include the cyclocondensation of acetonitrile derivatives such as cyanoacetate ester, cyanoacetamide, or malononitrile with an appropriate α,β-unsaturated carbonyl or 1,3-dicarbonyl substrate.…”
