Prognostic value of tumor-infiltrating lymphocytes in melanoma: a systematic review and meta-analysis

Abstract: Tumor-infiltrating lymphocytes (TILs) are associated with prognosis in various tumors. However, it remains controversial whether the presence of TILs is related to an improved prognosis in melanoma. This meta-analysis confirmed the favorable prognostic role of the CD3+, CD4+, CD8+, FOXP3+, and CD20 + TILs in the overall survival of melanoma patients and found an association between the TILs present and improved overall survival. Additionally, subgroup analysis demonstrated that brisk TILs were obviously associ… Show more

“…T-cell infiltration was associated with a better prognosis in all three of the tumors we examined. This was consistent with prior reports in melanoma and HNSCC 19,60 , but to our knowledge this is a novel finding in GBM that was likely missed by previous studies because T-cells are so rare in GBM.…”

“…During the past two decades numerous studies have revealed interactions between malignant cells and the stroma that promote diverse tumor functions including angiogenesis 4,5 , metastasis 6 , and immunosuppression 7,8 . Stromal cells differ between patients and tumor types [9][10][11][12][13][14][15] and the abundance of certain stromal cell populations are used in the clinic as biomarkers [16][17][18][19] and therapeutic targets [20][21][22][23][24][25] . These studies motivate direct measurements of cell types and the interactions between them in human subjects.…”

“…We did not observe similar effects of protein expression in our second validation cohort, which might be due to much smaller sample size. It is difficult to exclude the possibility that a significant component of the MX2 gene expression signal in the Leeds Melanoma Cohort is derived from TILs which themselves are a favorable prognostic marker in melanoma (Fu et al, ). The expression of MX2 in tumor cells did, however, correlate with the amount of immune cell infiltration implying that IFN secretion by TILs leads to induction of MX2 and other IFN‐dependent genes like MX1 .…”

“…Patients with tumors that are poorly oxygenated have an increased risk of mortality (22). In addition, the presence of tumor-infiltrated lymphocytes (TILs) can be used as a relevant prognostic indicator (23)(24)(25). A notorious hallmark associated with BCa intratumoral oxygen deprivation is alteration in the cancer cell survival metabolism to function through the hypoxia-inducible factor-1α (HIF-1α) regulatory pathway (26)(27)(28).…”
