Prognostic value of the PIK3CA, AKT, and PTEN mutations in oral squamous cell carcinoma: literature review

Abstract: Over 260,000 (2013) new oral squamous cell carcinoma (OSCC) cases are reported annually worldwide. Despite development in OSCC management, the outcome is still unsatisfactory. Identification of new molecular markers may be of use in prevention, prognosis, and choice of an appropriate therapy. The intracellular molecular signalling pathway of phosphatidyl-inositol-3-kinase is involved in the process of cell growth, differentiation, migration, and survival. The main components of this pathway: PIK3CA (phosphatidy… Show more

“…In the study of liver cancer, it was found that the full-length apoptosis-inducing factor mitochondrial-associated 1 ( AIFM1 ) (∼67 kDa) was cleaved at its N-terminus to produce truncated AIFM1 (∼57 kDa), which was not dependent on caspase Enzymes induce apoptosis, overexpression of full-length AIFM1 can induce caspase-dependent apoptosis and inhibit cell growth of liver cancer cells, our data reveal the potential role of rAd- AIFM1 in HCC gene therapy ( 31 ). Studies have shown that PIK3CA is a prognostic factor for oral cancer, and studies have also shown that PIK3CA is involved in the metabolic mechanism of breast cancer ( 32 , 33 ). Inhibition of PIK3CA can cause tumor regression in nude mice ( 34 ).…”

“…Oral carcinogenesis involves the accumulation of various genetic aberrations, including DNA mutation, epigenetic changes (DNA methylation) and chromosomal aberrations. More recently, NOTCH1, PIK3CA, AKT, PTEN and TRIM21 genomic changes have been identified as actionable genetic changes in oral cancers [5][6][7]. While chemotherapeutic regimens (cisplatin with or without fluorouracil and docetaxel) are most commonly used for the treatment of oral squamous cell carcinoma (OSCC), patients with prolonged chemotherapy develop a relapse due to chemoresistance [8].…”

“…Carcinomas of the tongue and floor of the oral cavity are frequently locally advanced at the time of the first presentation and show a worse prognosis than other oral cancers. Nevertheless, other factors related to tumor biology and its immune microenvironment may also influence the prognosis or be the targets for personalized therapy [ 2 , 3 , 4 ].…”
