Prognostic Significance of CSF-1R Expression in Early Invasive Breast Cancer

Abstract: Colony-stimulating factor-1 receptor (CSF-1R) signaling promotes an immune suppressive microenvironment enriched in M2 macrophages. Given that CSF-1R inhibitors are under investigation in clinical trials, including in breast cancer, CSF-1R expression and association with immune biomarkers could identify patients who derive greater benefit from combination with immunotherapies. TIMER2.0 and bc-GenExMiner v4.7 were used to assess the correlation of CSF1R mRNA with immune infiltrates and prognosis. Following a pr… Show more

“…CSF-1 also promotes immunosuppression through stimulating the differentiation of MDSCs and the selective activation of TAMs expressing CSF-1R and MHC-II. Immune cell infiltration with high expression of CSF-1R in the TME is usually associated with immune dysfunction and enhanced immune resistance, such as poor prognosis in ovarian cancer, breast cancer, endometrial cancer, PDAC, and lymphoma [ 129 132 ]. Blocking the CSF1/CSF-1R axis is the most established method for reducing the infiltration of TAMs.…”

“…In breast carcinoma cells, CSF-1R has been demonstrated as being functional and implicated in various biological effects, such as proliferation, invasion, and epithelial-mesenchymal transition [21,25,[49][50][51][52][53][54][55]. Moreover, its expression is correlated with poor clinical outcomes [22,56]. Thus, the role of the CSF-1/CSF-1R axis in supporting the growth and motility of several solid tumors has been well documented [57].…”

“…CSF-1R is also expressed in microglia and neural progenitor cells participating in central nervous system development [19]. CSF-1 and its receptor play a role in normal breast tissue during puberty, pregnancy, and lactation [13,20] and in breast carcinogenesis [21,22].…”

“…Among them, fibroblasts were derived from primitive mesenchymal cells and had been reported to interact with cancer cells, [41,42] so they were used as normal control cells. In addition, CSF-1R overexpression had been reported around osteosarcoma, [43] breast and lung cancer tissues, [44,45] so their corresponding three cell lines were used to evaluate the biological activity of compound P1. The results of the MTT experiment were shown in Figure 3.…”
