Prognostic potential of detection of WT1 mRNA level in peripheral blood in adult acute myeloid leukemia

Abstract: We retrospectively analyzed the potential of Wilms' tumor gene 1 (WT1) mRNA levels in peripheral blood for predicting the prognosis of 50 patients with AML. After achieving complete remission (CR), 34 patients (69.4%) were determined to be positive and 15 (30.6%) were negative for WT1. The relapse rate of the positive and negative patients was 73.5% and 40.0% (p = 0.02), respectively. After consolidation therapy, only 15 patients (32.6%) were positive and 31 (67.4%) were negative for WT1. Althoug… Show more

“…WT1, the gene mutated in Wilms' tumor, was found expressed in acute myeloid leukemia (AML), and the level of its expression was reported to be associated with minimal residual disease of AML after chemotherapy (9,10). Since WT1 level is very high at the time of AML diagnosis, PB is widely used for this purpose, though BM samples usually show about 10 times higher WT1 level than PB samples.…”

“…RNA was extracted from leukocytes in the PB and from nucleated cells in the BM using an RNeasy Mini Kit (Qiagen, Valencia, CA, USA), and WT1 mRNA was measured using a WT1 mRNA Assay Kit (Otsuka Pharmaceutical Co., Ltd., Tokyo, Japan) [11]. cDNA was synthesized from 1 µg of extracted RNA by reverse transcription using the random hexamer method.…”

“…Wilms' tumor 1 (WT1) is expressed in high frequency in the PB and BM of many patients with acute myeloid leukemia (AML) [20,21]. The development of in vitro WT1 messenger RNA (mRNA) diagnostics has resulted in its wide use in clinical practice as an MRD monitoring marker.…”
