Prognostic and predictive value of long non-coding RNA GAS5 and mircoRNA-221 in colorectal cancer and their effects on colorectal cancer cell proliferation, migration and invasion

Abstract: The results revealed that LncRNA GAS5 was upregulated while the miR-221 was downregulated in the tissues, plasma and exosomes of patients with CRC. The results of ROC showed that the expressions of both lncRNA GAS5 and miR-221 in the tissues, plasma and exosomes had diagnostic value in CRC. While the LncRNA GAS5 expression in tissues, plasma and exosomes were associated with the tumor node metastasis (TNM) stage, Dukes stage, lymph node metastasis (LNM), local recurrence rate and distant metastasis rate, the M… Show more

“…43 Reduced GAS5 facilitated colon cancer cell migration, proliferation and invasion. 44 Downregulated GAS5 also promoted the cell cycle in the G0/ G1 stage and prevented apoptosis. Increased GAS5 was an independent marker of a longer overall survival and better prognosis.…”

“…Yuan et al found that co‐overexpressed GAS5 and SNORD44 could significantly repress tumor growth and they could induce cancer cells apoptosis in both vivo and vitro . The overexpression of GAS5 could suppress colon cancer cells proliferation and promote apoptosis by inhibiting the expression of MIR‐182‐5p and MIR‐221 , but upregulating FOXO3a …”

“…Nevertheless, in a recent observation, it was demonstrated that lncRNA GAS5 conduces to lymphatic metastasis of CRC (19). Additionally, using qRT-PCR, Liu et al detected upregulation of lncRNA GAS5 and downregulation of miR-221 in CRC (20). Surprisingly, the level of lncRNA GAS5 is also related to clinical and pathological features (20).…”

“…Additionally, using qRT-PCR, Liu et al detected upregulation of lncRNA GAS5 and downregulation of miR-221 in CRC (20). Surprisingly, the level of lncRNA GAS5 is also related to clinical and pathological features (20). Therefore, the mechanism of lncRNA GAS5 in CRC warrants further study and confirmation.…”

“…In addition, microarray results indicated that the expression of the lncRNA GAS5 was decreased in SKBR-3/Tr cells and breast cancer tissues from trastuzumab-treated patients, suggesting that GAS5 may serve as a novel prognostic marker and candidate drug target for HER2-positive breast cancer (57). Collectively, the prognostic values of these lncRNAs for patients with cancer were assessed by meta-analysis, which may facilitate improvements in the diagnosis and prognosis of cancer treatment (58)(59)(60). Although the majority of pioneering studies remain in the early stage, they still encourage further examination of the diagnostics and therapeutic opportunities of lncRNAs in cancer.…”
