Background. Chromobox protein homolog 8 (CBX8), a transcriptional repressor, participates in many biological processes in various carcinomas. Cell differentiation, aging, and cell cycle progression are examples of such processes. It is critical to investigate CBX8 expression and its relationship with clinicopathological characteristics in liver hepatocellular carcinoma (LIHC), kidney renal clear cell carcinoma (KIRC), and ovarian cancer (OV) to investigate CBX8’s potential diagnostic and prognostic values. Methods. TCGA and CPTAC databases were used to compare the data between cancer and matched normal tissues on RNA and protein expression profiles and their relevant clinical information to determine the relationship between CBX8 and clinicopathological features. Kaplan–Meier analyses were used to assess CBX8 relationship’s with disease-free survival (DFS), relapse-free survival (RFS), and overall survival (OS). The multivariate Cox regression analysis was used to identify independent risk factors which affect prognosis. DNA methylation and genetic changes and their impact on prognoses were evaluated by cBioPortal and MethSurv websites. Spearman’s correlation was used to determine the relationship of CBX8 expression with somatic mutation. Tumor immune estimation resource (TIMER) was adopted to investigate the relationship between CBX8 and immune cell infiltration (ICI). CBX8-relevant genes and proteins are analyzed by EnhancedVolcano and STRING databases. The gene set enrichment analysis (GSEA) was performed to investigate the potential functions of CBX8. Results. CBX8 RNA and protein overexpression were confirmed in LIHC, KIRC, and OV ( p < 0.05 ). High CBX8 was significantly related to the clinical features and poor prognoses. The CBX8 genetic alteration rate was 3%. DNA methylation was also associated with prognoses. CBX8 closely interacted with ICI, TMB, MSI, purity, and ploidy. GO analyses revealed that CBX8-associated genes were enriched in biological processes, cell cycle regulation, and molecular functions. KEGG analyses exhibited that CBX8 was gathered in signaling pathway regulation. GSEA revealed that cell cycle, DNA replication, and Wnt signaling pathways were differentially enriched in the high CBX8 expression group. Conclusions. CBX8 could be a potential diagnostic and prognostic biomarker for LIHC, KIRC, and OV cancers.

Chromobox proteins in cancer: Multifaceted functions and strategies for modulation (Review)

Chromobox (CBX) proteins are important epigenetic regulatory proteins and are widely involved in biological processes, such as embryonic development, the maintenance of stem cell characteristics and the regulation of cell proliferation and apoptosis. Disorder and dysfunction of CBXs in cancer usually lead to the blockade or ectoptic activation of developmental pathways, promoting the occurrence, development and progression of cancer. In the present review, the characteristics and functions of CBXs were first introduced. Subsequently, the expression of CBXs in cancers and the relationship between CBXs and clinical characteristics (mainly cancer grade, stage, metastasis and relapse) and prognosis were discussed. Finally, it was described how CBXs regulate cell proliferation and self-renewal, apoptosis and the acquisition of malignant phenotypes, such as invasion, migration and chemoresistance, through mechanisms involving epigenetic modification, nuclear translocation, noncoding RNA interactions, transcriptional regulation, posttranslational modifications, protein-protein interactions, signal transduction and metabolic reprogramming. The study also focused on cancer therapies targeting CBXs. The present review provides new insight and a comprehensive basis for follow-up research on CBXs and cancer. Contents1. Introduction 2. Objective Examining the potential mechanism underlying the effect of galangin combined with oxaliplatin on the invasion and proliferation of colorectal cancer. Methods The effects of galangin combined with oxaliplatin on colorectal cancer (CRC) cell invasion and proliferation were assessed using CCK8 and Transwell assays. Using WB and qPCR, the expression of CBX3 in several CRC cell lines was identified. The levels of CBX3 in normal colorectal tissues and CRC tissues were compared using HPA and GEPIA, and the impact of CBX3 on the prognosis of CRC patients was investigated using GEPIA. To determine whether galangin targets CBX3 to influence the growth and invasion of CRC cells, cell assays were employed. The levels of ROS in HCT116 were measured by immunofluorescence following galangin treatment. The impact of CBX3 on immune cell infiltration in CRC was examined using TIMER. GEPIA produced a list of CBX3-related genes, and R was utilized to perform GO/KEGG enrichment analysis. Results We found that galangin combined with oxaliplatin inhibited the proliferation and invasion of CRC cells and down-regulated the expression of CBX3, which was rescued by overexpression of CBX3. DFS was shorter in those with elevated CBX3 expression. The level of ROS in HCT116 was significantly increased after galangin treatment. The invasion of CD8 + T cells and macrophages in the immunological microenvironment of CRC is favorably linked with increased expression of CBX3. Nuclear chromatin, histone binding, and covalent chromatin modification are enriched regions in CBX3 and related genes. Conclusion Galangin combine with oxaliplatin can inhibit the proliferation and invasion of colorectal cancer cells by down-regulating the expression of CBX3, and CBX3 is highly expressed in colorectal cancer and is related to short DFS.
