Progesterone Receptor-Mediated Regulation of N-Acetylneuraminate Pyruvate Lyase (NPL) in Mouse Uterine Luminal Epithelium and Nonessential Role of NPL in Uterine Function

Abstract: N-acetylneuraminate pyruvate lyase (NPL) catalyzes N-acetylneuraminic acid, the predominant sialic acid. Microarray analysis of the periimplantation mouse uterine luminal epithelium (LE) revealed Npl being the most downregulated (35×) gene in the LE upon embryo implantation. In natural pregnant mouse uterus, Npl expression increased 56× from gestation day 0.5 (D0.5) to D2.5. In ovariectomized mouse uterus, Npl was significantly upregulated by progesterone (P4) but downregulated by 17β-estradiol (E2). Progester… Show more

“…To determine the effects of steroid hormones on SR-B1 expression, immature female rats (n = 30) were randomly divided into six groups, and each rat received subcutaneously (s.c.) injections of sesame oil 200 L (C), P 4 , E 2 , E 2 plus P 4 , E 2 plus ICI-182780 or P 4 plus RU486, respectively, according to earlier studies (Ahn et al, 2011;Gangula et al, 2001;Jung et al, 2011;Xiao et al, 2013). The P 4 (2 mg/kg BW), E 2 (20 g/kg BW), ER antagonist ICI-182780 (500 g/kg BW) and PR antagonist RU486 (400 g/kg BW) were dissolved in 200 L of sesame oil, respectively.…”

“…Among the most upregulated and downregulated protein-coding genes (Fig. 1C), Atp6v0d2 (ATPase H + transporting V0 subunit D2) [5,6] , Cyp26a1 (Cytochrome P450 family 26 subfamily A member 1) [7] , Gjb2 (gap junction protein beta 2) [8] , Npl (N-acetylneuraminate pyruvate lyase) [6,9] , and Fxyd4 (FXYD domain containing ion transport regulator 4) [6] are specifically or mainly ( Cyp26a1 ) expressed in the LE. The upregulation or downregulation of these genes in the pregnant uteri (compared to the pseudo-pregnant uteri) on D3 at 22 hours (Fig.…”
