Profiling of miRNAs in porcine Sertoli cells

Abstract: Background: Sertoli cells (SCs) create a specialized environment to support and dictate spermatogenesis. MicroRNAs (miRNAs), a kind of~22 nt small noncoding RNAs, have been reported to be highly abundant in mouse SCs and play critical roles in spermatogenesis. However, the miRNAs of porcine SCs remain largely unknown. Methods: We isolated porcine SCs and conducted small RNA sequencing. By comparing miRNAs in germ cells, we systematically analyzed the miRNA expression pattern of porcine SCs. We screened the hig… Show more

“…RNA-seq data investigation shows that miR-24-3p exhibits a developmentalstage-specific expression profile in porcine testicular tissues [33,34]. In addition, miR-24-3p is also cloned from the mice Sertoli cells [35], and it also shows a higher expression in porcine Sertoli cells than in other spermatogenic cell types [36]. However, there is no evidence to explore the regulatory role of miR-24-3p in porcine Sertoli cell proliferation.…”

“…The production of the nutrient has been likened to lactation in mammals, and hence, it has been called crop milk [ 39 ]; meanwhile, the pigeon crop undergoes significant changes to the tissue structure during lactation [ 17 ]. miRNAs have demonstrated to play crucial functions in lipid metabolism, adipose tissue development and immune response [ 40 , 41 , 42 ]. In the present study, we found that over 40% of the small RNAs in pigeon crop were miRNAs; a total of 71 miRNAs were identified as significantly differentially expressed.…”

“…Data on miRNA in relation to testicular toxicity in minipigs are very limited or absent. However, in pigs, miRNA expression profiles from Sertoli cells have been assessed by sRNA-seq (Chen et al 2020). Mature Sertoli cells were isolated from 5-month-old pigs, and the Sertoli cell RNA was further analyzed by sRNA-seq.…”
