Arbuscular mycorrhizal fungi (AMF) are promoted as biofertilizers for sustainable agriculture. So far, most researchers have investigated the effects of AMF on plant growth under highly controlled conditions with sterilized soil, soil substrates or soils with low available P or low inoculum potential. However, it is still poorly documented whether inoculated AMF can successfully establish in field soils with native AMF communities and enhance plant growth. We inoculated grassland microcosms planted with a grass-clover mixture (Lolium multiflorum and Trifolium pratense) with the arbuscular mycorrhizal fungus Rhizoglomus irregulare. The microcosms were filled with eight different unsterilized field soils that varied greatly in soil type and chemical characteristics and indigenous AMF communities. We tested whether inoculation with AMF enhanced plant biomass and R. irregulare abundance using a species specific qPCR. Inoculation increased the abundance of R. irregulare in all soils, irrespective of soil P availability, the initial abundance of R. irregulare or the abundance of native AM fungal communities. AMF inoculation had no effect on the grass but significantly enhanced clover yield in five out of eight field soils. The results demonstrate that AMF inoculation can be successful, even when soil P availability is high and native AMF communities are abundant.

Arbuscular mycorrhizal (AM) fungi are often applied as bio-inoculants due to their plant growth promoting benefits. The objectives of the present work were to maximize the shelf life of monoxenically produced carrier-based AM fungal bio-inocula of Rhizoglomus intraradices and Funneliformis mosseae. Shelf life of in vitro produced inoculum was studied by assessing the infectivity potential of in vitro produced inocula in an optimum carrier formulation (vermiculite: cow dung powder: wood powder: wood ash in the ratio of 20:8:2:1) during storage at three different temperatures viz., 4 °C, 25 °C and room temperature (RT). The re-germination potential of in vitro produced spores from carrier-based inocula to in vitro conditions was also examined. The in vitro produced inocula stored at 25 °C remained viable up to 6 mo in the organic carrier formulation. 100 % germination was recorded when the spores of both AM species were cultured back to in vitro conditions indicating high viability, and efficiency of the carrier formulation in maintaining vigour of in vitro produced propagules.

Plants are meta-organisms that are associated with complex microbiomes. Many of the microorganisms that reside on plant surfaces (epiphytes) or within plant tissues (endophytes) do not cause any plant diseases but often contribute significantly to the nutrient supply of their host plant and can help the plant to overcome a variety of biotic or abiotic stresses. The yield potential of any plant depends not only on successful plant traits that improve, for example, the adaptation to low input conditions or other stressful environments but also on the plant microbiome and its potential to promote plant growth under these conditions. There is a growing interest to unravel the mechanisms underlying these beneficial plant microbe interactions because the activities of these microbial communities are of critical importance for plant growth under abiotic and biotic stresses and could lead to the development of novel strategies to improve yields and stress resistances of agronomically important crops. In this chapter, we summarize our current understanding of the beneficial interactions of soybean plants with arbuscular mycorrhizal fungi, nitrogen-fixing rhizobia, and fungal and bacterial endophytes and identify major knowledge gaps that need to be filled to use beneficial microbes to their full potential.
