Production of phosphatase by microorganisms isolated from discolored painted walls in a typical tropical environment: a Non-Parametric analysis

Abstract: The enzymatic activities occurring on painted walls in a tropical environment have become critical given the rate of aesthetic biodeterioration of such walls. Pigmented heterotrophic microorganisms isolated from discolored painted walls in Lagos, Nigeria were screened for their phosphatase activities. The strains were characterized by morphological methods and 16S rDNA analysis as belonging to the genera Aspergillus, Meyerozyma, Candida, Fusarium, Cerrena and Pseudomonas. The sequences were submitted to NCBI G… Show more

“…S32 isolated from an alfalfa rhizosphere in a heavy metal-contaminated reclamation area in Shanxi Province, China was capable of producing ACP with a maximum activity of 69.4 U L −1 , which was 2.98 times lower than the activity obtained in the current study. Furthermore, the optimal ACP production of P. aeruginosa CH01 (18.4 U L −1 ), was attained at 96 h, which was 11.25 times less than the maximal activity obtained by the strain under study 11 . On the other hand, ACP was produced in large quantities by Klebsiella and Morganella , as well as Providencia stuartii .…”

“…However, there are so few bacteria in the biosphere that can solubilize phosphate. There is tremendous competition for phosphate among microorganisms, even though many species produce phosphatases and have elaborate mechanisms for regulating their production and activity 11 . Therefore, industries are still seeking novel bacterial species capable of producing industrial enzymes to meet the current ACP enzyme demand.…”

“…Amplification of the 16S rDNA gene, sequencing, and similarity. The genomic DNA of the potential ALP-producing isolate was extracted by the salting-out method 33 . The PCR reaction and sequencing were executed according to the method detailed by Abdelgalil et al 34 .…”

“…Upon performing quantitative screening, the medium was inoculated with 2% of 12 h activated old preculture of the promising isolate and incubated in a rotary shaker incubator (200 rpm) at 30 °C for 72 h. The supernatant with soluble target enzyme was obtained by centrifugation of the bacterial culture cellular pellet at 4 °C, 6000 rpm for 15 min and used to estimate ALP activity. Spectrophotometric assays for estimating ALP activity were conducted by following the absorbance change at 405 nm with a molar extinction coefficient (ε) of 18,000 M −1 cm −1 accompanying the hydrolysis of the chromogenic substrate (ρNPP) into ρ-nitrophenol, as reported by Obidi et al 32 . For the standard assay, ρNPP was added to the reaction mixture of 0.1 M modified universal buffer pH 10.0 to a final concentration of 10 mM.…”
