Production of monoclonal antibodies against serotype strains of Pseudomonas aeruginosa

Abstract: A panel of 22 monoclonal antibodies against 8 of the 17 International Antigenic Typing Scheme (IATS) serotypes of Pseudomonas aeruginosa was produced. The antibodies were characterized for cross-reactivities, isotypes, titers, and epitope specificities. The results complemented those of our previous study and marked the completion of a set of monoclonal antibodies for serotyping P. aeruginosa.

“…Clinical strains of P. aeruginosa isolated from patients with CF and sera from patients were obtained from the State Serum Institute, Rigshospitalet, Copenhagen, Denmark. These strains were typed by using both the Pseudomonas typing sera (Difco Laboratories, Detroit, Mich.) and a set of 17 serotype-specific MAbs produced in our laboratory (16,17). A set of serial isolates, strains 49 to 58, was obtained from one patient with CF over a 1-month period, with samples being taken every 2 to 3 days.…”

“…In our laboratory, we use the IATS (149,212) designations for 20 reference serotype strains. MAb which differentiate between the first 17 of the 20 IATS serotypes have been generated (132,133), while the remaining 3 IATS serotypes can be distinguished by using adsorbed polyclonal sera. In a recent review (212), P. aeruginosa B-band LPS was divided into 31 chemotypes that include the 20 IATS serotypes and subtypes (i.e., strains that react with an MAb for a particular IATS serotype but have additional epitopes).…”

“…Various efforts have been made to serotype l? aeruginosa (5,12,21,24,25,39,43). Different serological typing schemes based on 0antigens describe from five ( 3 ) up to 25 0groups (45) of the bacteria, and a distinct 0serotype in one system may contain two or more type-specific epitopes in other typing systems (19,27).…”
