Production of Migration Inhibitory Factor in Inbred Rats

Abstract: Strains of genetically inbred rats representative of the known Ag-B groups produced migration inhibitory factor in response to immunization with live bacillus Calmette-Guerin bacilli and intracardiac challenge with old tuberculin; the assays were performed by both in vitro and in vivo methods. None of the strains immunized in this fashion developed delayed hypersensitivity skin reactions to purified protein derivative.

“…The purpose of the current study was t o define whether human B cell lines express F c IgM receptors. Human hematopoietic cell lines represent a steadily proliferating resource of homogeneous cells expressing different surface receptors according t o their origin [5] and the procedure used for establishing the lines [6,7]. Most long-term human lymphoid cell lines show B cell characteristics, express Epstein Barr (EB) viral genome, and certain B cell lines secrete Ig of a specific class.…”

“…Guided by successful production of MIF by a number of inbred rat strains other than Lewis [6], and access to unpublished data from this same laboratory [Chen and Gill, unpublished observations], we inject ed Lewis rats with live BCG. Supernatants of purified protein derivative-stimulated peripheral blood leuco cytes (PBL), splenocytes, and lymph node cells (LNC) were tested for MIF activity over an arbitrarily chosen 50-fold concentration range.…”

“…The serum to be assayed for SIF (Salvin et al, 1973;Ruscetti et al, 1976) was stored frozen until used. Then a 0.6 ml aliquot was pipetted into 30 ml tissue culture flasks (Fischer Scientific Co., Blawnox, PA) which had 2.4 ml of 0.5% agar containing medium 199 and 25 mM Hepes buffer, pH 7.0.…”

“…Each serum was assayed in quadruplicate and the data for all of the animals in each experimental group were averaged to give the mean value for the percentage inhibition. The experimental variation in the assay method had been established previously (Ruscetti et al, 1976): <15% inhibition was not significant and 21 5% was significant. These limiting values are consistent with the variation in the assay results with sera from syngeneic pregnancies (Table 1).…”
