Production of Microbial Polyester by Fermentation of Recombinant Microorganisms

Abstract: Polyhydroxyalkanoates (PHAs) can be produced from renewable sources and are biodegradable with similar material properties and processibility to conventional plastic materials. With recent advances in our understanding of the biochemistry and genetics of PHA biosynthesis and cloning of the PHA biosynthesis genes from a number of different bacteria, many different recombinant bacteria have been developed to improve PHA production for commercial applications. For enhancing PHA synthetic capacity, homologous or h… Show more

“…After 16-18 h of growth in an orbital shaker (3.5 × g) at 30 or 35 °C (depending on the temperature of the subsequent cultivation, which was the same), an aliquot (calculated OD 600 = 0.1) was utilized as inoculum for the main cultivation in the same synthetic medium, as described below. (Lee and Choi, 2001). All medium components, except the carbon source, and thiamine were sterilized by autoclaving at 121 °C for 15 min.…”

“…Decades of biopolyester research were dedicated to understanding the production of these biopolymers and investigating the isolated crystallized biopolymers, their properties and potential applications (Brandl et al 1990, Lee & Choi 2001, Zinn et al 2001. However, the spherical polyester inclusions are now increasingly considered as bioparticles, which can be applied in medicine (Rehm 2006).…”

“…This was not only done in Arabidopsis thaliana [Poirier et al, 1992] but also in agriculturally relevant crops being already cultivated on large acreage for production of various food, feed and chemical compounds ( table 2 ). Successful expression of the R. eutropha PHA biosynthesis genes encouraged researchers also to use PHA biosynthesis genes from other microorganisms [Lee and Choi, 2001]. …”
