Production of Lycopene in the Non-Carotenoid-Producing Yeast Yarrowia lipolytica

Abstract: The codon-optimized genes crtB and crtI of Pantoea ananatis were expressed in Yarrowia lipolytica under the control of the TEF1 promoter of Y. lipolytica. Additionally, the rate-limiting genes for isoprenoid biosynthesis in Y. lipolytica, GGS1 and HMG1, were overexpressed to increase the production of lycopene. All of the genes were also expressed in a Y. lipolytica strain with POX1 to POX6 and GUT2 deleted, which led to an increase in the size of lipid bodies and a further increase in lycopene production. Lyc… Show more

“…[58] In addition, crtE gene or HMG1 gene encoding HMGR was overexpressed, enhancing lycopene production. [41,58,59] In other study, since the cytosol FPP pool partly flows into mitochondria, amorpha-4,11-diene synthase (ADS) was fused with mitochondrion-targeting signal peptide (mtADS) to utilize mitochondrial FPP as well ( Figure 6). [60] By coexpressing mtADS with the native ADS, amorphadiene production was greatly increased.…”

“…The purification of pigments from the yeast Phaffia rhodozyma was performed by chromatography using acetone and identification was performed by electronic absorption mass spectroscopy, confirming the synthesis of astaxanthin by the yeast (Johnson and Lewis, 1979). The production of lycopene by Yarrowia lipolytica was confirmed using HPLC with various compositions of the mobile phase, water, methanol, acetonitrile and ethyl acetate (Matthäus et al, 2014).…”
