Production of Immunoabsorbent Nanoparticles by Displaying Single‐Domain Protein A on Potato Virus X

Abstract: The combination of antibodies with nanoparticles provides wide-ranging applications in biosensing. While several covalent presentation strategies have been established, there is need for alternative, non-covalent methods to provide a routine for scalable nanomanufacturing. We report the multivalent presentation of the B domain of Staphylococcus aureus protein A (SpAB) on potato virus X (PVX) nanoparticles. Three different synthetic strategies were used to obtain chimeric PVX(SpAB) filaments. The protein A frag… Show more

“…The study elucidated that amino acid content, mainly tryptophan, and isoelectric point could drastically alter the trafficking of the PVX through plants. For biosensing and diagnostic applications, PVX was genetically altered to display protein A fragments from Staphylococcus aureus [87]. The protein A coated PVX was then readily functionalized with IgG and was applied in biosensing of plant viruses [88].…”

“…As another approach, it was recently demonstrated that functionalization of PVX-based nanoparticles with protein A fragments can be used to display whole antibody molecules by using protein A's property of binding to the Fc region of the heavy chain of IgGs. 85 The PVX particles could then be used as a plug-and-play system for the display of a variety of antibodies, with a level of orientational control not achievable with chemical conjugation.…”

“…For PVX, an alternative methodology to display a target protein is the insertion of the FMDV 2A sequence between the target gene and the cp , hence a ribosomal skip occurs during translation, leading to the expression of the CP fusion protein, free target protein as well as wild-type CP (Cruz et al, 1996; Donnelly et al, 2001a,b). The FMDV 2A sequence has previously been used in PVX vectors to avoid unfavorable limitations of CP fusions (Cruz et al, 1996; Lee et al, 2014; Shukla et al, 2014; Uhde-Holzem et al, 2015), but to our knowledge it has not yet been used in a TMV CP fusion protein.…”
