Production of Glycolic Acid by Chemolithotrophic Iron- and Sulfur-Oxidizing Bacteria and Its Role in Delineating and Sustaining Acidophilic Sulfide Mineral-Oxidizing Consortia

Abstract: Glycolic acid was detected as an exudate in actively growing cultures of three chemolithotrophic acidophiles that are important in biomining operations, Leptospirillum ferriphilum, Acidithiobacillus (At.) ferrooxidans, and At. caldus. Although similar concentrations of glycolic acid were found in all cases, the concentrations corresponded to ca. 24% of the total dissolved organic carbon (DOC) in cultures of L. ferriphilum but only ca. 5% of the total DOC in cultures of the two Acidithiobacillus spp. Rapid acid… Show more

“…Generally, these organics are detrimental to chemolithotrophic autotrophic microorganisms, especially Leptospirillum spp. (Ñancucheo and Johnson, 2010;Okibe and Johnson, 2004;Zhang et al, 2014). Moreover, the Table 1 Specific primers for the quantification of 16s rRNA gene copies of each strain in the mixed consortium.…”

“…Concentrations of metals (other than chromium) in samples that had been filtered through 0.2 μm pore-sized polycarbonate filters were determined by high pressure ion chromatography (HPIC; [14]). Chromium concentrations were determined by atomic adsorption spectrophotometry.…”

“…Parallel analysis of proteins and carbohydrates was carried out using harvested biomass of "F. myxofaciens" P3G. To identify the sugars present in EPS from both native acid streamers and a pure culture of "F. myxofaciens" P3G, EPS were first extracted and hydrolyzed (20) and monosaccharides were identified using ion chromatography (21).…”
