Production of chiral R‐3‐hydroxyalkanoic acids and R‐3‐hydroxyalkanoic acid methylesters via hydrolytic degradation of polyhydroxyalkanoate synthesized by pseudomonads

Abstract: A novel and efficient method for the production of enantiomericaly pure R-3-hydroxyalkanoic acids and R-3-hydroxyalkanoic acid methylesters was developed. The described method is based on hydrolysis of poly(hydroxyalkanoate) copolymers synthesized by Pseudomonas putida. The polymer was isolated via solvent recovery and hydrolyzed by acid methanolysis. The obtained 3-hydroxyalkanoic acid methylester mixture was distilled into several fractions with an overall yield of 96.6% (w/w). Gas chromatography-mass spectr… Show more

“…The PhaZ pathway has reportedly achieved up to 7.3 g L -1 (R)-3HB production in a shake flask culture (Shiraki et al 2006). This method has an advantage over (R)-3HB synthesis by the acidic hydrolysis of P(3HB) that generates acid derivatives (de Roo et al 2002, Shiraki et al 2006. However, the PhaZ pathway requires a two step fermentation, accumulation of the P(3HB) polymer and also its degradation.…”

“…the fraction that passed through the filter) were finally resuspended in chloroform, methanolysed and analysed by GC. A series of controls were used to optimise the performance of the filter columns: pure mcl-PHA control consisting of polymer isolated from P. putida KT2442; pure monomer consisting of 3-(R)-hydroxyoctanoate obtained as described elsewhere (de Roo et al 2002) and kindly provided by Guy de Roo; a mix of polymer and monomer.…”

“…PHA is also considered as pharmaceutically-active compound and currently investigated as potential anti-HIV drugs, anti-cancer drugs, antibiotics, etc. [7,8]. The production of various types of PHA material, their properties and downstream applications was recently reviewed by Philip et al [9], Olivera et al [10], Chen [11], and Rai et al [7].…”
