Production of Basic Fibroblast Growth Factor‐like Factor by Cultured Human Cholangiocellular Carcinoma Cells

Abstract: An extract of cultured human cholangiocellular carcinoma cells (HuCC‐T1) was found to contain high mitogenic activity for BALB/c3T3 cells. The growth factor eliciting most of the mitogenic activity was purified and concluded to be identical with basic fibroblast growth factor (bFGF)‐like factor on the basis of its molecular weight and heparin‐Sepharose elution profile, and the results of immunoblotting and radioimmunoassay. HuCC‐T1 cells also secreted bFGF‐like factor into serum‐free medium. A combination of i… Show more

“…3A). This preparation maximally stimulated DNA synthesis in BALB/c3T3 cells at 90 pg/ml (5 pM), indicating that mitogenic activity of the recombinant human bFGF is equivalent to that of bFGF purified from bovine brain (41). When the Pgk-1 gene was transcribed in nuclear extracts of Ehrlich ascites tumor cells, exogenous bFGF greatly inhibited transcription from two different DNA constructs (Fig.…”

“…The primary actions of bFGF are generally considered to be either of the following: (1) autocrine action in Which tumor cells generate bFGF and prompt growth of tumor cells themselves [13,14]; or (2) paracrine action in which growth of the peripheral vascular endothelial cells is prompted [12,15]. Because bFGF is an unstable substance and its half-life in blood is short (it can be expressed in seconds), the endocrine action of bFGF is an exceptional one in which, similar to insulin-like growth factors and hepatocellular proliferation factors, bFGF is generated and secreted into the blood by cells in the other organs, is transported to the blood flow, and prompts proliferation of the target tumor cells or endothelial cells in the vessels in the periphery of tumors [16].…”

“…The cells were changed to serum-free medium in the absence or presence of 40 pm recombinant human TGFb 1 (R&D System), and incubated for 20 h. DNA synthesis was measured by the incorporation of 1 mCi/ml [ 3 H]thymidine (Amersham Pharmacia Biotech, Buckinghamshire, UK) into 5% trichloroacetic acid-precipitable material after a 4 h pulse, as previously described (Matsuzaki et al, 1990). Experimental and control values were compared using the unpaired Student's t-test and ANOVA; Po0.05 was considered to be significant.…”
