Production and Purification of d -Aminoacylase from Alcaligenes denitrificans and Taxonomic Study of the Strain

Abstract: A D-aminoacylase-producing microorganism, strain DA181, isolated from soil was identified as Alcaligenes denitrifcans subsp. denitrificans. This strain produced about 29,300 units (micromoles of product formed per hour) of D-aminoacylase and 2,300 units of L-aminoacylase per gram of cells (wet weight) when cultivated in a medium containing 1% N-acetyl-DL-leucine as the carbon source. The D-aminoacylase was purified 345-fold. The specific activity of the purified enzyme was 108,600 units per mg of protein when … Show more

“…In our previous article, the production and characterization of the D-aminoacylase produced by Alcaligenes denitrificans subsp. denitrificans DA181 was reported (13). This enzyme exhibited not only high stereospecificity but also high specific activity in comparison with the D-aminoacylases from Pseudomonas and Streptomyces spp.…”

“…or Streptomyces olivaceus can be used. [13][14][15] Watanabe et al 9 also developed D-pipecolic acid synthesis via hydrolysis of Boc-D-amino-6-bromohexanoic acid ethyl ester to Boc-D-2-amino-6-bromohexanoic acid followed by removal of the Boc group and consequent cyclization by addition of triethylamine as reported in the case of L-pipecolic acid. Many studies were performed on amino acid racemases; nevertheless, no pipecolic acid racemase has been hypothesized to be involved in D-pipecolic acid catabolism so far.…”

“…The mutations were confirmed by DNA sequencing. D-Aminoacylase activity was measured using N-acetyl-D-methionine as substrate coupled to a D-amino acid oxidase enzyme assay as previously described (Tsai et al 1988). For the inhibition assay, each thiol-specific reagent was incubated with the enzyme for 20…”
