Production and localization of restrictocin in Aspergillus restrictus

Abstract: The production and secretion of restrictwin (a cytotoxin that cleaves ribosomal RNA) by cultures of the fungus Aspergillus restrictus was investigated. Previous studies have indicated that restrictwin production in liquid culture coincides with the appearance of differentiated cell structures. A study of the correlation between the appearance of differentiated structures and restrictwin production was conducted with A. restrictus grown on agar medium. Restrictwin was found to be associated with the cell mass o… Show more

“…These results showed a significant decrease in feeding on the first of these two fungi when conidia were starting to develop, while no change in the insect behavior was observed against maturing A. nidulans. The period of not consumption was coincident with a dramatic build up of restrictocin upon the surfaces of conidia and phialides (Brandhorst and Kenealy, 1992) suggesting that it could deter insects from feeding until conidia were fully formed. Once the spores were mature, the ribotoxin levels would decrease allowing insects to carry spores to new locations for spreading the fungus.…”

“…Intemalisation of this protein can be also achieved by the construction of immunoconjugates, prepared by the attachment of the ribotoxin to monoclonal antibodies specific of receptors present on the surface of the target cells (Conde et al, 1989;Wawrzynczak et al, 1991). The occurrence of this ribosome inactivating protein in the course of an Aspergillus infection and its presence on the conidial surface has prompted several groups to consider this molecule as a potential virulence factor in aspergillosis Arruda et al, 1992;Brandhorst and Kenealy, 1992).…”
