Production and Characterization of Recombinant Active Mouse Gelatinase B from Eukaryotic Cells and in vivo Effects after Intravenous Administration

Abstract: Gelatinase B is a matrix metalloproteinase involved in tissue remodelling. When mouse cells are triggered in vitro with interleukin-I, bacterial endotoxin, virus-mimicking double-stranded RNA or cytokine inducers, they produce gelatinase B. To test the effects of gelatinase B in vivo, the enzyme was expressed in Chinese hamster ovary (CHO) cells. Hybrid genomic DNA-cDNA constructs under the control of two constitutive viral promoters were generated by PCR-mediated exon amplification. In vitro transcription and… Show more

“…3A). We ascribed this activity to MMP-9 due to its proximity to human MMP-9; its apparent molecular mass in zymograms corresponded to murine MMP-9, which has been estimated at 105 to 110 kDa (25,41), and the presence of a comigrating MMP-9-specific signal on a mouse-specific MMP-9 Western blot (see Fig. 4, below).…”

Indeed, it has been proposed that an important step in leucocyte exit from the bone marrow is the degradation of the extracellular matrix within the bone marrow stroma (Opdenakker et al, 1998).…”

“…These data are apparently at odds with a previous study reporting that i.v. injection of recombinant MMP-9 elicits a blood leucocytosis (Masure et al, 1997). However, it is possible that the leucocytosis generated by the i.v.…”

“…and S.F., unpublished observations, January 2003). Furthermore, administration of recombinant MMP-9 to rabbits induces a rapid peripheral leukocytosis, 67 although HSCs were not quantitated. It is likely that in GRO␤/GRO␤ T -mobilized mice, elevation of marrow MMP-9 that degrades matrix and releases HSCs, plus disruption of endothelial junctions by plasma MMP-9, coordinately result in HSC transmigration.…”
